【摘要】：目的研究基質(zhì)交感分子1(stim1)和鈣離子釋放激活鈣離子通道蛋白1(orai1)在哮喘氣道平滑肌細胞中表達水平的變化。方法選取40只清潔級SD大鼠隨機分為對照組和哮喘組,其中哮喘組用雞卵清蛋白(OVA)激發(fā)制備大鼠哮喘模型(n=20),對照組用生理鹽水代替(n=20)。肺組織病理切片HE染色判斷哮喘模型是否建立成功,分析支氣管肺泡灌洗液(BALF)中的細胞總數(shù)及分類計數(shù),應(yīng)用免疫組化技術(shù)及彩色病理圖文分析系統(tǒng)測定stim1和orai1蛋白陽性細胞平均光密度值。結(jié)果肺組織病理切片HE染色鏡下觀察呈現(xiàn)大量炎細胞浸潤,支氣管黏膜腫脹,支氣管腔內(nèi)可見黏液栓等,證實哮喘模型建立成功。哮喘組中大鼠BALF中細胞總數(shù)、中性粒細胞、嗜酸性粒細胞計數(shù)明顯高于對照組,差異有統(tǒng)計學(xué)意義(P0.01)。哮喘組大鼠支氣管平滑肌表達orai1蛋白陽性細胞平均光密度值明顯高于對照組(P0.05)。哮喘組和對照組大鼠支氣管平滑肌表達stim1蛋白陽性細胞的平均光密度值無明顯差異(P0.05)。結(jié)論 orai1蛋白在哮喘大鼠氣道平滑肌細胞中表達變化較stim1蛋白明顯,對支氣管平滑肌調(diào)節(jié)作用可能更重要。
[Abstract]:Objective to investigate the expression of matrix sympathetic molecule 1stim1 and Ca2 + releasing activated Ca2 + channel protein 1 in asthmatic airway smooth muscle cells (ASSMC). Methods Forty SD rats of clean grade were randomly divided into control group and asthmatic group. The asthmatic group was stimulated with ovalbumin (OVA) and the control group was treated with normal saline instead of OVA. Lung tissue sections were stained with HE to determine whether the asthma model was successful or not, and the total number of cells in bronchoalveolar lavage fluid (BALF) and its classification count were analyzed. The average optical density of stim1 and orai1 protein positive cells was measured by immunohistochemical technique and color pathological image and text analysis system. Results A large number of inflammatory cells, swelling of bronchial mucosa and mucus embolus in bronchial cavity were observed under HE staining in pathological sections of lung tissue. It was proved that asthma model was established successfully. The number of BALF cells, neutrophils and eosinophils in asthma group was significantly higher than that in control group (P 0.01). The average optical density of orai1 protein positive cells in bronchial smooth muscle in asthma group was significantly higher than that in control group (P 0.05). There was no significant difference in optical density between asthma group and control group in the expression of stim1 protein positive cells in bronchial smooth muscle. Conclusion the expression of orai1 protein in airway smooth muscle cells of asthmatic rats is more obvious than that of stim1 protein, which may play a more important role in regulating bronchial smooth muscle.
