枸杞多糖對COPD患者缺氧誘導(dǎo)因子1α抑制作用的研究

【摘要】:目的明確枸杞多糖(LBP)對慢性阻塞性肺疾病(COPD)穩(wěn)定期患者外周靜脈血中缺氧誘導(dǎo)因子1α的抑制作用并探討枸杞多糖濃度與HIF-1α的關(guān)系。方法1.收集101名入組對象,其中包括51例COPD穩(wěn)定期患者,50例健康者,所有患者均有完整的臨床資料。2.檢測COPD穩(wěn)定期組和健康對照組的血清HIF-1α指標(biāo),通過t檢驗(yàn)進(jìn)行組間比較,判斷HIF-1α在COPD穩(wěn)定期患者的表達(dá)水平。3.檢測COPD穩(wěn)定期組服用不同劑量枸杞多糖前、后血清HIF-1α指標(biāo),通過重復(fù)測量資料的方差分析,分析HIF-1α水平與劑量濃度及觀察時(shí)間的關(guān)系。4.檢測COPD患者動脈血?dú)饧胺喂δ苤笜?biāo)。結(jié)果1.COPD穩(wěn)定期患者外周靜脈血中HIF-1α顯著高于健康者,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。2.COPD穩(wěn)定期患者服用枸杞多糖2周后檢測外周靜脈血中HIF-1α表達(dá)水平較服用枸杞多糖前下降,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。3.本實(shí)驗(yàn)中觀察COPD穩(wěn)定期患者服用不同劑量LBP后HIF-1α的變化趨勢發(fā)現(xiàn)低劑量組下降趨勢明顯高于中、高劑量組。4.COPD穩(wěn)定期組患者HIF-1α與PO2呈負(fù)相關(guān)(r=-0.49,P0.05),與PCO2呈正相關(guān)(r=0.283,P0.01);HIF-1α與患者的FEV1%呈負(fù)相關(guān)(r=-0.42,P0.05),與FEV1/FVC呈負(fù)相關(guān)(r=-0.54,P0.05)。5.服用枸杞多糖2周后,13例患者胸悶、氣短癥狀改善,有效率30.23%;7例患者咳痰不利癥狀改善,有效率25.92%;7例患者體力增強(qiáng)、感冒次數(shù)減少,有效率17.95%;2例患者精神狀態(tài)改善明顯,有效率18.18%。結(jié)論1.COPD穩(wěn)定期患者血清中HIF-1α表達(dá)水平較健康者明顯升高。2.COPD穩(wěn)定期患者血清中HIF-1α表達(dá)水平與機(jī)體缺氧、二氧化碳潴留及氣流受限程度相關(guān)。3.LBP對COPD穩(wěn)定期患者血清中HIF-1α表達(dá)具有抑制作用,本實(shí)驗(yàn)中低劑量LBP對HIF-1α的抑制作用優(yōu)于中、高劑量組。4.LBP可以改善COPD患者的部分臨床癥狀,提高其生活質(zhì)量。
[Abstract]:Objective to investigate the inhibitory effect of Lycium barbarum polysaccharides (LBP) on hypoxia inducible factor-1 偽 (HIF-1 偽) in peripheral venous blood of patients with chronic obstructive pulmonary disease (COPD) at stable stage and to explore the relationship between LBPs concentration and HIF-1 偽. Method 1. 101 subjects were collected, including 51 stable COPD patients and 50 healthy subjects, all of whom had complete clinical data. 2. 2. Serum HIF-1 偽 levels were measured in stable COPD group and healthy control group. The expression level of HIF-1 偽 in stable COPD patients was determined by t test. Serum HIF-1 偽 was measured before and after different doses of Lycium barbarum polysaccharides in stable COPD group. The relationship between HIF-1 偽 level and dose concentration and observation time was analyzed by variance analysis of repeated measurement data. Arterial blood gas and pulmonary function were measured in COPD patients. Results 1. The level of HIF-1 偽 in peripheral venous blood of stable COPD patients was significantly higher than that of healthy controls. The difference was statistically significant (P 0.05). 2. The expression of HIF-1 偽 in peripheral venous blood of stable COPD patients was significantly lower than that before taking Lycium barbarum polysaccharides for 2 weeks. The difference was statistically significant (P 0.05). In this experiment, the trend of HIF-1 偽 in stable COPD patients with different doses of LBP was observed. The decrease trend of HIF-1 偽 in low dose group was significantly higher than that in middle dose group. 4. There was a negative correlation between HIF-1 偽 and PO2 in stable COPD group, a positive correlation between HIF-1 偽 and PCO2, a negative correlation between HIF-1 偽 and FEV1%, and a negative correlation between HIF-1 偽 and FEV1 / FVC, a negative correlation between HIF-1 偽 and FEV1 / FVC, a negative correlation between HIF-1 偽 and FEV1 / FVC, a negative correlation between HIF-1 偽 and FEV1 / FVC, a negative correlation between HIF-1 偽 and PCO2, a negative correlation between HIF-1 偽 and FEV1 / FVC, and a negative correlation with FEV1 / FVC, respectively. After taking Lycium barbarum polysaccharides for 2 weeks, 13 cases had chest tightness, the symptoms of shortness of breath were improved, the effective rate was 30.23%, the effective rate was 25.92%, the physical strength of 7 cases was enhanced, the number of colds was decreased, and the effective rate was 17.95%. The efficiency is 18.18. Conclusion 1. The expression of HIF-1 偽 in serum of stable COPD patients was significantly higher than that of healthy controls. 2. The expression of HIF-1 偽 in serum of stable COPD patients was significantly higher than that of healthy controls. The effect of low dose LBP on HIF-1 偽 expression in stable COPD patients was better than that in middle and low dose group. 4.LBP in high dose group could improve some clinical symptoms of COPD patients. Improve their quality of life.
