慢性阻塞性肺病患者冷敏受體高表達及其臨床意義

【摘要】:目的探討瞬時受體電位蛋白-8(TRPM8)在慢性阻塞性肺病(COPD)患者與正常人氣道黏膜上皮組織的表達差異及其信號傳導機制。方法免疫組化法觀察TRPM8蛋白在氣道黏膜上皮分布特點及表達差異,RT-PCR及Western bolt檢測TRPM8 mRNA及其蛋白表達。體外培養(yǎng)16HBE細胞(n=5),分為對照組(37℃)、冷刺激組、冷刺激+BCTC組、冷刺激+轉染TRPM8 shRNA組、冷刺激+轉染對照組,以18℃冷空氣刺激細胞,激光共聚焦檢測胞內Ca2+相對濃度。結果 TRPM8蛋白主要分布于支氣管黏膜上皮基底細胞或小顆粒細胞的細胞膜,COPD組累積吸光度/面積比值、TRPM8 mRNA及其蛋白表達量顯著高于對照組(P0.05)。冷刺激組的Ca2+濃度顯著高于對照組(P0.05),BCTC、轉染TRPM8 shRNA組較前者明顯下降(P0.05)。結論 COPD患者氣道黏膜上皮TRPM8蛋白高表達可能是其對冷空氣敏感的重要原因之一。
[Abstract]:Objective to investigate the expression and signal transduction mechanism of transient receptor potential protein (TRPM8) in epithelial tissue of airway mucosa in patients with chronic obstructive pulmonary disease (COPD) and normal subjects.
