【摘要】：探討菠蘿蛋白酶誘導(dǎo)小鼠急性過(guò)敏性氣道炎癥中2型固有淋巴細(xì)胞(group 2innate lymphoid cell,ILC2)及相關(guān)細(xì)胞因子的變化及意義。對(duì)未致敏BALB/c雌性小鼠在第1天和第3天給予菠蘿蛋白酶滴鼻,ELISA檢測(cè)12h內(nèi)小鼠肺組織勻漿上清中IL-33變化,5d后處死小鼠,肺組織切片病理觀察,瑞氏染色計(jì)數(shù)BALF中嗜酸性粒細(xì)胞含量,流式檢測(cè)肺組織中ILC2數(shù)量變化,ELISA檢測(cè)BALF中IL-5、IL-13水平。結(jié)果顯示,菠蘿蛋白酶誘導(dǎo)氣道上皮細(xì)胞產(chǎn)生IL-33,3h時(shí)開(kāi)始,6h時(shí)達(dá)高峰;菠蘿蛋白酶滴鼻小鼠肺組織切片顯示大量炎性細(xì)胞浸潤(rùn)和杯狀細(xì)胞黏液分泌增加,BALF中嗜酸性粒細(xì)胞比例增加,IL-5和IL-13含量增加,流式檢測(cè)肺組織中ILC2數(shù)量增多。研究表明,菠蘿蛋白酶刺激小鼠氣道上皮細(xì)胞釋放IL-33,引起小鼠肺ILC2活化增殖并分泌大量Th2型細(xì)胞因子,引起嗜酸性粒細(xì)胞浸潤(rùn)等急性過(guò)敏性氣道炎癥表現(xiàn),提示ILC2促進(jìn)早期急性過(guò)敏性氣道炎癥的發(fā)生。
[Abstract]:To investigate the changes and significance of type 2 inherent lymphocyte group 2innate lymphoid cell2 (ILC2) and related cytokines in acute allergic airway inflammation induced by bromelain in mice. The unsensitized BALB/c female mice were killed on the 1st and 3rd day by using bromelain nasal drip Elisa to detect the changes of IL-33 in the lung tissue supernatant of the mice within 12 hours. The eosinophilic granulocytes in BALF were counted by Rayleigh staining and the changes of ILC2 in lung tissue were detected by flow cytometry. IL-5 and IL-13 in BALF were detected by Elisa. The results showed that the production of IL-33 was induced by bromelain and reached the peak at the beginning of 6h. The lung tissue sections of mice fed with bromelain showed that a large number of inflammatory cells infiltrated and goblet cell mucus secretion increased. The percentage of eosinophils increased in BALF and the contents of IL-5 and IL-13 were increased. The number of ILC2 in lung tissue was increased by flow cytometry. The results showed that bromelain stimulated the release of IL-33 from mouse airway epithelial cells, induced the activation and proliferation of ILC2 and the secretion of a large number of Th2 cytokines in the lung of mice, and caused the infiltration of eosinophilic granulocytes and other acute allergic airway inflammation. The results suggest that ILC2 promotes the occurrence of early acute allergic airway inflammation.
