地塞米松對肺炎支原體感染大鼠血清及肺泡灌洗液中IL-12、IL-13的影響

【摘要】:目的研究地塞米松對肺炎支原體(MP)感染大鼠血清及肺泡灌洗液(BALF)中IL-12、IL-13的影響,探討其治療MP肺炎的作用機制。方法 100只SD大鼠隨機分為4組:阿奇霉素組、地塞米松組、聯(lián)合干預(yù)組和模型對照組,每組各25只。各組大鼠均予MP菌液(1×106CFU/ml)緩慢滴入大鼠鼻腔,共4 d,在最后1次接種后第2天開始干預(yù)。采用ELISA方法監(jiān)測干預(yù)后第3、5、8天大鼠血清及BALF液IL-12、IL-13水平,并對肺組織炎癥浸潤程度進行評分。結(jié)果在干預(yù)第3、5、8天,阿奇霉素組、地塞米松組、聯(lián)合干預(yù)組及模型對照組肺組織病理評分比較差異均有統(tǒng)計學(xué)意義(F=8.71~20.11,P均0.01),其中均以聯(lián)合干預(yù)組評分最低。在干預(yù)第3、5、8天,四組間血清及BALF液中IL-12、IL-13水平比較差異均有統(tǒng)計學(xué)意義(F=9.52~143.47,P均0.01);其中干預(yù)第3天,聯(lián)合干預(yù)組血清IL-12高于阿奇霉素組,而血清及BALF液IL-13均低于阿奇霉素組,差異均有統(tǒng)計學(xué)意義(P均0.05)。結(jié)論地塞米松可能協(xié)同阿奇霉素通過促進大鼠體內(nèi)IL-12分泌和抑制IL-13表達,減輕肺組織炎癥。
[Abstract]:Objective to study the effect of dexamethasone on IL-12 IL-13 in serum and alveolar lavage fluid (BALF) of rats infected with MP, and to explore the mechanism of the treatment of MP pneumonia. Methods 100 SD rats were randomly divided into 4 groups: azithromycin group, dexamethasone group, combined intervention group and model control group with 25 rats in each group. All the rats in each group were treated with 1 脳 10 6 CFU / ml MP bacteria solution, which was slowly dripped into the nasal cavity of the rats for 4 days, and the intervention began on the second day after the last inoculation. The levels of IL-12 IL-13 in serum and BALF fluid of rats were monitored by ELISA method on the 3rd day and 8th day after intervention, and the degree of inflammatory infiltration in lung tissue was evaluated. Results on the 5th day of intervention, the pathological scores of lung tissue in azithromycin group, dexamethasone group, combined intervention group and model control group were significantly different. On the 3rd day of intervention, the levels of IL-12 and IL-13 in serum and BALF solution were significantly different among the four groups (P < 0.01), and on the third day of intervention, the serum IL-12 of the combined intervention group was higher than that of the azithromycin group, while the serum and BALF solution IL-13 levels were lower than those of the azithromycin group. The difference was statistically significant (P < 0.05). Conclusion Dexamethasone combined with azithromycin may reduce lung inflammation by promoting IL-12 secretion and inhibiting IL-13 expression in rats.
