【摘要】：目的探索白細(xì)胞介素-25(IL-25)通過(guò)nuocyte細(xì)胞對(duì)哮喘小鼠氣道重塑的作用。方法 30只SPF級(jí)BALB/c雌性小鼠隨機(jī)分為對(duì)照小鼠組、哮喘小鼠組、抗IL-25小鼠組,每組10只,雞清卵蛋白(OVA)誘導(dǎo)哮喘模型。分離培養(yǎng)哮喘小鼠組nuocyte細(xì)胞,并分為對(duì)照細(xì)胞組、IL-25細(xì)胞組、抗IL-25細(xì)胞組。HE染色觀察肺組織變化,流式細(xì)胞法計(jì)數(shù)肺泡灌洗液(BALF)中nuocyte細(xì)胞,ELISA法檢測(cè)BALF及細(xì)胞上清液中IL-4、IL-13含量,免疫組化、Western blotting及RT-PCR法檢測(cè)α-平滑肌肌動(dòng)蛋白(α-SMA)、Ⅰ膠原蛋白(collagen-Ⅰ)在肺組織中的表達(dá)。結(jié)果與對(duì)照小鼠組相比,哮喘小鼠組BALF中IL-4、IL-13表達(dá)升高,nuocyte細(xì)胞增多,α-SMA和collagen-Ⅰ表達(dá)升高(P0.05),而抗IL-25小鼠組表達(dá)無(wú)明顯變化(P0.05);與對(duì)照細(xì)胞組相比,IL-25細(xì)胞組上清液IL-4、IL-13表達(dá)增高(P0.05),抗IL-25細(xì)胞組上清液IL-4、IL-13表達(dá)無(wú)明顯變化(P0.05)。結(jié)論IL-25可能通過(guò)nuocyte細(xì)胞促使TH2型細(xì)胞因子分泌,導(dǎo)致哮喘小鼠氣道重塑。
[Abstract]:Objective to investigate the effect of interleukin-25 (IL-25) on airway remodeling in asthmatic mice by nuocyte cells. Methods Thirty SPF grade BALB/c female mice were randomly divided into control group, asthma group and anti IL-25 group, each group with 10 ovalbumin (ovalbumin) to induce asthma model. Nuocyte cells of asthmatic mice were isolated and divided into control group and IL-25 cell group. The changes of lung tissue were observed by HE staining in anti IL-25 cell group. Nuocyte cells in alveolar lavage fluid (BALF) were counted by flow cytometry to detect the content of BALF and IL-4 IL-13 in supernatant. The expression of 偽 -smooth muscle actin (偽 -SMA-I) in lung tissue was detected by immunohistochemistry (Western blotting and RT-PCR).
