IL-25通過nuocyte細胞誘導哮喘小鼠氣道重塑

【摘要】:目的探索白細胞介素-25(IL-25)通過nuocyte細胞對哮喘小鼠氣道重塑的作用。方法 30只SPF級BALB/c雌性小鼠隨機分為對照小鼠組、哮喘小鼠組、抗IL-25小鼠組,每組10只,雞清卵蛋白(OVA)誘導哮喘模型。分離培養(yǎng)哮喘小鼠組nuocyte細胞,并分為對照細胞組、IL-25細胞組、抗IL-25細胞組。HE染色觀察肺組織變化,流式細胞法計數(shù)肺泡灌洗液(BALF)中nuocyte細胞,ELISA法檢測BALF及細胞上清液中IL-4、IL-13含量,免疫組化、Western blotting及RT-PCR法檢測α-平滑肌肌動蛋白(α-SMA)、Ⅰ膠原蛋白(collagen-Ⅰ)在肺組織中的表達。結(jié)果與對照小鼠組相比,哮喘小鼠組BALF中IL-4、IL-13表達升高,nuocyte細胞增多,α-SMA和collagen-Ⅰ表達升高(P0.05),而抗IL-25小鼠組表達無明顯變化(P0.05);與對照細胞組相比,IL-25細胞組上清液IL-4、IL-13表達增高(P0.05),抗IL-25細胞組上清液IL-4、IL-13表達無明顯變化(P0.05)。結(jié)論IL-25可能通過nuocyte細胞促使TH2型細胞因子分泌,導致哮喘小鼠氣道重塑。
[Abstract]:Objective to investigate the effect of interleukin-25 (IL-25) on airway remodeling in asthmatic mice by nuocyte cells. Methods Thirty SPF grade BALB/c female mice were randomly divided into control group, asthma group and anti IL-25 group, each group with 10 ovalbumin (ovalbumin) to induce asthma model. Nuocyte cells of asthmatic mice were isolated and divided into control group and IL-25 cell group. The changes of lung tissue were observed by HE staining in anti IL-25 cell group. Nuocyte cells in alveolar lavage fluid (BALF) were counted by flow cytometry to detect the content of BALF and IL-4 IL-13 in supernatant. The expression of 偽 -smooth muscle actin (偽 -SMA-I) in lung tissue was detected by immunohistochemistry (Western blotting and RT-PCR).
