【摘要】：目的觀察白介素22(IL-22)在哮喘模型中的作用,研究布地奈德對(duì)哮喘小鼠模型氣道炎癥及IL-22的調(diào)控作用。方法 24只BALB/c小鼠隨機(jī)分為對(duì)照組、哮喘模型組和布地奈德組3組,用卵清蛋白(OVA)致敏、激發(fā)小鼠以制備哮喘模型。小鼠肺組織進(jìn)行HE及AB-PAS染色,進(jìn)行氣道炎癥評(píng)分,ELISA法檢測(cè)3組小鼠肺泡灌洗液(BALF)中IL-22的水平,實(shí)時(shí)熒光定量PCR檢測(cè)小鼠肺組織中IL-22mRNA的表達(dá)水平。結(jié)果與對(duì)照組相比較,哮喘小鼠肺組織炎癥評(píng)分增加,BALF中IL-22水平增高,肺組織中IL-22mRNA表達(dá)水平升高,差異均具有統(tǒng)計(jì)學(xué)意義。給予布地奈德治療后,小鼠氣道炎癥評(píng)分及肺組織IL-22mRNA的表達(dá)水平均較哮喘組降低,差異具有統(tǒng)計(jì)學(xué)意義。結(jié)論布地奈德對(duì)哮喘氣道炎癥的治療作用與其抑制肺內(nèi)IL-22的表達(dá)和分泌有關(guān)。
[Abstract]:Objective to observe the role of interleukin 22 (IL 22) in asthma model and to study the effect of budesonide on airway inflammation and IL-22 in asthmatic mice. Methods Twenty-four BALB/c mice were randomly divided into three groups: control group, asthma model group and budesonide group. The mice were sensitized with ovalbumin (ovalbumin) to induce asthma model. The lung tissues of mice were stained with HE and AB-PAS. The level of IL-22 in alveolar lavage fluid (BALF) was detected by Elisa and the expression of IL-22mRNA in lung tissue was detected by real-time fluorescence quantitative PCR. Results compared with the control group, the inflammatory score of lung tissue in asthmatic mice increased the level of IL-22 and the expression of IL-22mRNA in lung tissue. The difference was statistically significant. After treatment with budesonide, the airway inflammation score and the expression of IL-22mRNA in lung tissue were significantly lower in mice than in asthma group. Conclusion the therapeutic effect of budesonide on asthmatic airway inflammation is related to its inhibition of IL-22 expression and secretion in the lung.
