PM2.5吸入對(duì)哮喘大鼠氣道炎癥的影響

【摘要】:目的:PM2.5對(duì)哮喘大鼠氣道炎癥和體重的影響進(jìn)行研究。方法:將124只雄性SD大鼠隨機(jī)分為4組:A(正常組)、B(哮喘未吸入PM2.5組)、C1(哮喘吸入低劑量PM2.5組)、C2(哮喘吸入高劑量PM2.5組)。模型大鼠給予卵清蛋白腹腔注射兩次致敏,2周后再用卵清蛋白連續(xù)霧化吸入14天,建立大鼠過敏性哮喘模型。PM2.5模型組,每周一至周三連續(xù)3天,每天1次,每次20分鐘超聲霧化吸入PM2.5。在吸入PM2.5一周、二周、四周、六周后觀察各組大鼠哮喘發(fā)作表現(xiàn)和體重情況。霧化結(jié)束1天后取材,進(jìn)行BALF白細(xì)胞計(jì)數(shù)和分類,ELISA試劑盒檢測(cè)血清總IgE,BALF中IL-6、IL-8含量。結(jié)果:1.成功建立SD大鼠哮喘模型。2.PM2.5對(duì)哮喘大鼠體重的影響B(tài)組大鼠在建模期、染毒一周和染毒二周時(shí)與A組比較有顯著差異,其中在建模期和染毒一周時(shí)差異非常顯著(P0.01),染毒二周時(shí)P0.05;C2組在建模期、染毒期各個(gè)時(shí)間段與A組比較均有顯著差異(P0.01),且在染毒四周、六周時(shí)與B組比較有顯著差異,染毒四周時(shí)差異更顯著(P0.01);C1組結(jié)果與C2組結(jié)果相似,兩組的差異僅在于后者染毒二周時(shí)與B組比較有統(tǒng)計(jì)學(xué)差異。3.PM2.5對(duì)哮喘大鼠BALF中白細(xì)胞計(jì)數(shù)和分類的影響染毒第一周,C1組、C2組BALF中白細(xì)胞總數(shù)均明顯低于B組(均p 0.01);C2組:嗜堿細(xì)胞百分比明顯低于A組、B組和C1組;淋巴細(xì)胞百分比明顯高于B組和A組;C1組:淋巴細(xì)胞百分比明顯高于B組。染毒第二周,C1組及C2組白細(xì)胞總數(shù)均明顯低于A組,白細(xì)胞分類各組間無統(tǒng)計(jì)學(xué)差異。染毒第四周,C1組中性粒細(xì)胞百分比明顯高于A組和B組;C1組和C2組:嗜酸細(xì)胞百分比明顯低于A組。染毒第六周,C2組中性粒細(xì)胞百分比明顯高于A組、B組和C1組。4.PM2.5對(duì)哮喘大鼠血清IgE、BALF中IL-6、IL-8含量的影響染毒第一周,BALF中IL-8含量C1組明顯低于A組和B組,C2組明顯低于B組,但明顯高于C1組。染毒第二周,血清IgE、BALF中IL-6、IL-8含量無統(tǒng)計(jì)學(xué)差異。染毒第四周,C1組BALF中IL-6含量明顯低于A組;IL-8含量C1組和C2組均明顯低于A組和B組。染毒第六周,C1組大鼠血清IgE含量出現(xiàn)明顯升高,明顯高于A組和B組;BALF中IL-6含量C1組明顯高于A組。不同染毒時(shí)間段血清IgE含量,C1組:染毒第四周明顯低于第二周,染毒第六周又明顯高于第一周和第四周;C2組:染毒第六周明顯高于第四周。結(jié)論:1.PM2.5能使哮喘大鼠體重明顯下降,染毒第六周,吸入PM2.5的哮喘大鼠體重也未能達(dá)到正常大鼠和哮喘大鼠體重水平。2.染毒第一周:PM2.5能加重哮喘大鼠氣道炎癥,并且與PM2.5劑量相關(guān),氣道炎癥以淋巴細(xì)胞反應(yīng)為主;染毒四周以后,PM2.5導(dǎo)致的氣道炎癥加重,以中性粒細(xì)胞反應(yīng)為主;整個(gè)染毒期間嗜酸細(xì)胞和嗜堿性粒細(xì)胞在氣道炎癥反應(yīng)中不起主要作用。3.PM2.5使哮喘大鼠BALF中IL-8、IL-6含量均出現(xiàn)明顯變化,尤其是吸入低劑量PM2.5組更明顯,并且變化幅度IL-8遠(yuǎn)大于IL-6,其出現(xiàn)變化時(shí)間也明顯早于IL-6。4.染毒第六周,PM2.5使哮喘大鼠血清IgE含量出現(xiàn)明顯增高,早期PM2.5吸入未啟動(dòng)IgE介導(dǎo)的哮喘炎癥反應(yīng)途徑。
[Abstract]:Objective: To study the effect of PM2.5 on airway inflammation and body weight in asthmatic rats. Methods: 124 male SD rats were randomly divided into 4 groups: A (normal group), B (asthma uninhaled PM2.5 group), C1 (asthma inhaled low dose PM2.5 group), C2 (asthma inhaled high dose PM2.5 group). Model rats were given ovalbumin intraperitoneal two times sensitization, and then ovalbumin after 2 weeks. The.PM2.5 model group was set up for 14 days after inhalation of the protein, and the model group was established for 3 days from Monday to Wednesday, 1 times a day. PM2.5. inhaled by ultrasonic atomization at 20 minutes each time was inhaled for one week, two weeks, four weeks, and six weeks to observe the symptoms and body weight of the rats in each group. After the atomization, the materials were obtained for 1 days and BALF white blood cells were obtained. Count and classify the total IgE of serum, IL-6 and IL-8 in BALF. Results: 1. the effect of.2.PM2.5 on the weight of asthma rat model of SD rats was successfully established in the modeling period of group B, and there was a significant difference between the group of B rats and the group of A for one week and two weeks, and the difference between the modeling period and the one week was very significant (P0.01). The effect of.3.PM2.5 on leukocyte count and classification of BALF in asthmatic rats for the first week, the total number of white blood cells in group C1 and C2 was significantly lower than that in group B (P 0.01); C2 group: the percentage of basophils was significantly lower than that of the A group, B group and C1 group; the percentage of lymphocytes was significantly higher than that in the B group and the group; the percentage of lymphocytes was significantly higher than that of the group. Second weeks, the total number of white blood cells in group C1 and C2 group was significantly lower than that of group A. There was no statistical difference between groups of white blood cells. The percentage of neutrophils in group C1 was significantly higher than that in group A and B group; the percentage of eosinophils in group C1 and C2 group was significantly lower than that in group A. The percentage of neutrophils in C2 group was significantly higher than that in A group. The effect of PM2.5 on the serum IgE, IL-6 and IL-8 content in the serum of the asthmatic rats was infected for the first week. The IL-8 content in C1 group was obviously lower than that of the A group and B group in the BALF group, and the C2 group was obviously lower than that of the B group, but the content of the serum was significantly higher than that in the group of B. The content of the serum was significantly lower than that in the group. The content of the serum was significantly lower than that in the group. Conclusion: 1.PM2.5 can reduce the weight of asthmatic rats significantly, and the body weight of asthmatic rats can not be reduced significantly. The body weight of the asthmatic rats inhaled PM2.5 can not reach the normal rats and the weight level.2. of the asthmatic rats for the first week. PM2.5 can aggravate the airway inflammation in the asthmatic rats, and it is related to the dose of PM2.5, the airway inflammation is drenched. After four weeks of exposure, PM2.5 resulted in increased airway inflammation and neutrophil reaction. During the whole exposure, eosinophils and basophils did not play a major role in airway inflammation,.3.PM2.5 made the BALF IL-8 and IL-6 content of the asthmatic rats significantly changed, especially in the low dose PM2.5 group. More obviously, the amplitude of IL-8 was much greater than that of IL-6, and its change time was obviously earlier than that of IL-6.4. for sixth weeks. PM2.5 increased the serum IgE content in the asthmatic rats, and the early PM2.5 inhalation did not start the IgE mediated asthma inflammatory response.
