【摘要】：目的: 探索通過向腹腔內(nèi)注射骨髓間充質(zhì)干細胞（BMSCs）培養(yǎng)液是否能夠改善支氣管哮喘小鼠肺部炎癥的病理反應的程度，能否下調(diào)肺泡灌洗液及血清中T淋巴細胞相關(guān)的細胞因子的水平。 方法:隨機將30只實驗小鼠分為三組，，每組10只： 1.對照組：實驗用藥物均用注射用生理鹽水代替（腹腔內(nèi)注射及霧化吸入藥物）。 2.哮喘組；制作哮喘小鼠動物模型，不予以干預。 3.治療組；在每次激發(fā)前的2小時將2ml的骨髓間充質(zhì)干細胞培養(yǎng)液向小鼠的腹腔內(nèi)注射。這樣一共激發(fā)三次，最后一次激發(fā)后48小時將實驗小鼠處死，采用病理方法分析收集到的小鼠的血清、肺泡灌洗液以及肺臟組織。 結(jié)果: 1.哮喘組小鼠的肺組織與正常的對照組相比，出現(xiàn)明顯的變態(tài)反應性炎癥反應（Underwood評分顯著升高，P0.01） 2.治療組的小鼠與哮喘組相比，肺部變態(tài)反應性炎癥反應顯著減輕（Underwood評分降低，P0.01） 3.哮喘組與對照組相比，小鼠肺泡灌洗液中細胞總數(shù)顯著增多（P0.01）。 4.治療組的小鼠與正常對照組相比，肺泡灌洗液中的細胞總數(shù)增多（P0.05）；而與哮喘組的小鼠相比，肺泡灌洗液中的細胞總數(shù)減少（P0.05）。 5.哮喘組小鼠與對照組相比，血清及肺泡灌洗液中IL-17水平顯著升高（P0.01）；BMSCs培養(yǎng)液治療組小鼠血清及肺泡灌洗液中IL-17水平比哮喘組顯著降低（P0.05）。 6. BABL/c小鼠的血清以及肺泡灌洗液中的IL-4水平，在正常對照組、哮喘組以及治療組中無統(tǒng)計學差別。 結(jié)論: 將BMSCs培養(yǎng)液注于哮喘小鼠腹腔內(nèi)，可以減輕其肺部炎性病理反應的程度，下調(diào)肺泡灌洗液及血清中T淋巴細胞相關(guān)的IL-17的水平。
[Abstract]:Objective:To explore whether intraperitoneal injection of bone marrow mesenchymal stem cells (BMSCs) can improve the degree of pulmonary inflammation in asthmatic mice and down-regulate the levels of cytokines associated with T lymphocytes in alveolar lavage fluid and serum.Methods: thirty experimental mice were randomly divided into three groups with 10 mice in each group.1.Control group: all the experimental drugs were replaced by normal saline (intraperitoneal injection and atomization inhalation).2.Asthma group; make asthma mice model, no intervention.3.In the treatment group, bone marrow mesenchymal stem cells (BMSCs) of 2ml were injected intraperitoneally into mice 2 hours before each challenge.The mice were killed 48 hours after the last stimulation. The collected serum, alveolar lavage fluid and lung tissue were analyzed by pathological method.Results:1.Compared with the normal control group, the lung tissue of the asthmatic mice showed obvious allergic inflammatory response, the Underwood score increased significantly (P 0.01).2.Compared with asthma group, lung allergic inflammation was significantly reduced in treatment group (P 0.01).3.Compared with the control group, the total number of cells in the alveolar lavage fluid increased significantly in the asthma group (P 0.01).4.Compared with the control group, the total number of cells in the alveolar lavage fluid increased in the treatment group (P 0.05) and decreased in the lung alveolar lavage fluid (P 0.05) compared with that in the asthma group.5.Compared with the control group, the level of IL-17 in serum and alveolar lavage fluid in asthma group was significantly higher than that in asthmatic group. The level of IL-17 in serum and alveolar lavage fluid in treatment group was significantly lower than that in asthma group.6.The levels of IL-4 in serum and alveolar lavage fluid of BABL/c mice were not significantly different in normal control group, asthma group and treatment group.Conclusion:Injecting BMSCs culture medium into the abdominal cavity of asthmatic mice could reduce the degree of pulmonary inflammatory pathological reaction and down-regulate the level of T lymphocyte related IL-17 in alveolar lavage fluid and serum.
