血管內(nèi)皮生長(zhǎng)因子受體抑制劑SU5614對(duì)哮喘模型小鼠氣道炎癥的影響

【摘要】:目的觀察血管內(nèi)皮生長(zhǎng)因子(VEGF)受體抑制劑SU5614對(duì)甲苯二異氰酸酯(TDI)誘發(fā)哮喘模型小鼠VEGF表達(dá)、氣道反應(yīng)性和氣道炎癥的影響。方法 BALB/C小鼠30只,隨機(jī)分為3組(n=10):對(duì)照組、模型組和SU5614組。采用TDI作為致敏原制備小鼠哮喘模型。SU5614組:SU5614溶解于二甲基亞砜(DMSO)中,于激發(fā)1 h后按照2.5 mg/kg給予腹腔注射,每日1次,連續(xù)3 d;模型組:只造模,不用藥;對(duì)照組:未造模小鼠。觀察各組小鼠VEGF的表達(dá)情況、氣道反應(yīng)性的變化,以及支氣管肺泡灌洗液中細(xì)胞總數(shù)和白細(xì)胞分類(lèi)的變化。結(jié)果 Western blotting和免疫組織化學(xué)分析結(jié)果顯示:模型組VEGF表達(dá)明顯增加,SU5614組則明顯降低。模型組氣道反應(yīng)性明顯高于對(duì)照組,SU5614組則明顯低于模型組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);模型組小鼠支氣管肺泡灌洗液中細(xì)胞總數(shù)以及中性粒細(xì)胞、巨噬細(xì)胞、淋巴細(xì)胞和嗜酸性粒細(xì)胞數(shù)明顯高于對(duì)照組,SU5614組則明顯低于模型組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 SU5614可抑制支氣管肺泡灌洗液中VEGF的表達(dá),降低氣道高反應(yīng)性,減輕氣道炎癥。
[Abstract]:Objective to observe the effects of vascular endothelial growth factor (VEGF) receptor inhibitor SU5614 on the expression of VEGF, airway reactivity and airway inflammation in asthmatic mice induced by toluene diisocyanate (TDI). Methods Thirty BALB/C mice were randomly divided into 3 groups: control group. Model group and SU5614 group. Mice asthma model was made by using TDI as allergen. Group SU5614 was dissolved in dimethyl sulfoxide (DMSO) and injected intraperitoneally according to 2.5 mg/kg once a day for 3 days. Control group: no model mice. The expression of VEGF and the changes of airway reactivity in each group were observed. Results the results of Western blotting and immunohistochemical analysis showed that the expression of VEGF in the model group was significantly increased and the airway reactivity in the model group was significantly lower than that in the su5614 group. It was significantly higher than that of the control group (Su5614) and lower than that of the model group (P < 0.05). The number of neutrophils, macrophages, lymphocytes and eosinophilic granulocytes in the bronchoalveolar lavage fluid of the model group was significantly higher than that in the control group (P < 0.05), and the number of neutrophils, macrophages, lymphocytes and eosinophils in the model group was significantly lower than that in the control group. Conclusion SU5614 can inhibit the expression of VEGF in bronchoalveolar lavage fluid, reduce airway hyperresponsiveness and reduce airway inflammation.
