褪黑素對矽肺大鼠肺纖維化的影響

【摘要】:[目的]研究褪黑素(melatonin,MT)對矽肺大鼠肺纖維化的影響及其可能機制。[方法]將72只SPF級SD雄性大鼠,隨機分為4組(對照組、MT組、染塵組及MT+染塵組),每組18只。對照組及MT組采用氣管插管灌注法,一次性氣管內(nèi)注入1 m L生理鹽水,其他兩組采用同樣方法注入50 mg/m L的二氧化硅懸液1 m L。每天給予MT組、MT+染塵組腹腔注射MT(10 mg/kg),而染塵組及對照組給予腹腔注射同體積生理鹽水。每組分別于染毒后第7、14、28天隨機處死6只大鼠。觀察大鼠的肺臟器系數(shù)、肺組織形態(tài),測定肺組織羥脯氨酸(Hyp)含量、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、肺組織核轉(zhuǎn)錄因子-κB(NF-κB)的表達。[結果]MT+染塵組大鼠肺泡炎癥細胞浸潤、充血水腫、肺間質(zhì)纖維化的程度較染塵組相比有明顯改善;與同期染塵組大鼠相比,MT+染塵組大鼠肺臟器系數(shù)、Hyp含量、MDA含量,SOD活性增加,差異均有統(tǒng)計學意義(均P0.05)。Western blot檢測結果顯示,MT+染塵組大鼠肺組織NF-κB蛋白表達量(0.36±0.02)低于染塵組(0.48±0.01),差異有統(tǒng)計學意義(P0.05)。[結論]褪黑素對矽肺大鼠肺纖維化有保護作用,其作用機制可能是通過改善機體氧化/抗氧化系統(tǒng)的失衡、抗炎,從而減少纖維化的形成。
[Abstract]:[objective] to study the effect of melatonin on pulmonary fibrosis in silicosis rats and its possible mechanism. [methods] Seventy-two SPF male SD rats were randomly divided into 4 groups (control group). There were 18 rats in each group in Dust group and MT group. The control group and MT group were infused with tracheal intubation, and 1 mL saline was injected into the trachea at one time. The other two groups were injected with 50 mg/m / L silica suspension 1 mL / day. The MT group was given intraperitoneal injection of MT(10 mg / kg of MT(10 mg / kg per day, while the dust-infected group and the control group were given intraperitoneal injection of normal saline of the same volume. Six rats were killed at random on the 28th day of the 7th day, and the lung organ coefficients of the rats were observed. The lung tissue morphology, the content of hydroxyproline, the activity of superoxide dismutase (SOD), the content of malondialdehyde (MDA) and the expression of nuclear transcription factor 魏 B (NF- 魏 NF- 魏 B) in lung tissue were measured. [results] the alveolar inflammatory cells infiltrated and hyperemia and edema were observed in MT dusty rats. The degree of pulmonary interstitial fibrosis was significantly improved than that of dust-exposed group, and the content of Hyp content and MDA content and SOD activity in lung tissue were increased in MT dust-exposed group compared with that in dust-exposed group. The expression of NF- 魏 B protein in lung tissue of rats exposed to MT dust was 0.36 鹵0.02, which was significantly lower than that of dust-exposed group (0.48 鹵0.01). [conclusion] melatonin has protective effect on pulmonary fibrosis in silicosis rats. The mechanism may be to reduce the formation of fibrosis by improving the imbalance of oxidation / antioxidant system and anti-inflammation.
