【摘要】：目的解整合素-金屬蛋白酶33(ADAM33)是哮喘重要的易感基因之一,與哮喘發(fā)病和氣道高反應(yīng)性密切相關(guān)。文中評(píng)估ADAM33在不同嚴(yán)重程度哮喘患者血清中表達(dá)水平及其與氣道慢性炎癥的關(guān)系。方法回顧性分析2015年5月1日至2016年5月1日期間于南京總醫(yī)院就診的輕-中度哮喘(n=54)、重度哮喘(n=35)和對(duì)照組(n=30)患者,分別檢測(cè)血清總Ig E水平、外周血嗜酸性粒細(xì)胞(EOS)計(jì)數(shù)和血清中ADAM33、IL-4和IL-13表達(dá)水平,并進(jìn)行相關(guān)性分析。結(jié)果輕-中度哮喘組、重度哮喘組和對(duì)照組患者血清中ADAM33表達(dá)分別為(23.8±6.21)、(64.8±12.8)和(18.3±4.49)pg/m L,重度哮喘組和輕-中度哮喘組血清ADAM33表達(dá)均顯著高于健康對(duì)照組,且重度哮喘組血清ADAM33表達(dá)也同樣高于輕-中度哮喘組(P0.05)。相關(guān)分析結(jié)果顯示,ADAM33表達(dá)與IL-4和IL-13呈正相關(guān)性(r=0.79、r=0.81,P0.05),而與EOS計(jì)數(shù)和血清總Ig E無(wú)相關(guān)性(r=0.54、r=0.46,P0.05)。結(jié)論 ADAM33在不同嚴(yán)重程度哮喘患者血清中的升高可反映哮喘的嚴(yán)重程度。血清IL-4和IL-13表達(dá)水平與ADAM33的正相關(guān)提示ADAM33的表達(dá)水平可能受到Th2類細(xì)胞因子的調(diào)控。
[Abstract]:Objective integrin-metalloproteinase-33 (ADAM33) is one of the important susceptibility genes to asthma. The expression of ADAM33 in serum of asthmatic patients with different severity of asthma and its relationship with chronic airway inflammation were evaluated. Methods from May 1st 2015 to 2016, we retrospectively analyzed the relationship between the expression of ADAM33 and chronic airway inflammation. From 1st to 1st, patients with mild to moderate asthma (54 cases) and severe asthma patients (35%) and control group (30 cases) with mild to moderate asthma were treated at Nanjing General Hospital in 1st. The levels of serum IgE, eosinophil eosinophils (EOS) in peripheral blood and the expression of ADAM33, IL-4 and IL-13 in serum were measured, and the correlation analysis was carried out. The expression of ADAM33 in serum of patients with severe asthma and control group was 23.8 鹵6.21, 64.8 鹵12.8) and 18.3 鹵4.49 PG / mL, respectively. The expression of serum ADAM33 in severe asthma group and mild to moderate asthma group was significantly higher than that in healthy control group. The expression of serum ADAM33 in severe asthma group was also higher than that in mild to moderate asthma group (P 0.05). The results of correlation analysis showed that the expression of ADAM33 was positively correlated with IL-4 and IL-13, but had no correlation with EOS count and serum IgE count. Conclusion ADAM33 is of different severity. The positive correlation between the expression of IL-4 and IL-13 and ADAM33 suggests that the expression of ADAM33 may be regulated by Th2 cytokines.
