Th2細胞和相關(guān)細胞因子在繼發(fā)性間質(zhì)性肺炎中的表達及意義

【摘要】:目的觀察皮下注射博來霉素致間質(zhì)性肺炎小鼠模型中Th2細胞及相關(guān)細胞因子的變化,探討博來霉素致繼發(fā)性間質(zhì)性肺炎的可能機制。 方法將20只Balb/c小鼠隨機分為2組:對照組(A組):皮下注射磷酸鹽緩沖液,10只;模型組(B組):皮下注射博來霉素,10只。Masson和HE染色后觀察小鼠肺部病理的改變,評估纖維化和炎癥程度。流式細胞術(shù)檢測外周血和肺組織Th2細胞的比例,免疫熒光定量PCR法檢測肺組織TGF-β1mRNA和IL-10mRNA的表達量,ELISA法檢測外周血LTGF-β1和IL-10的含量,并分析這些指標之間的相關(guān)性。 結(jié)果(1)B組小鼠肺纖維化評分[3.5(3,5)]、肺部炎癥評分[2(2,3)]均高于A組[0.5(0,1)和0(0,1)],(均P0.05);(2)外周血和肺組織Th2細胞比例B組[(3.32±2.13)%和(5.72±3.37)%]明顯高于A組[(1.21±0.63)%和(1.51±0.34)%];(3)外周血TGF-β1含量B組[(1426.31±137.08)pg/ml]明顯高于A組[(1138.44±92.61)pg/ml],外周血IL-10含量B組[(122.09±47.62) pg/ml]明顯高于A組[(76.82±25.74)pg/ml](均P0.05);(4)肺組織TGF-β1mRNA和IL-10mRNAB組[0.2100(0.0418,0.4700)和0.0261(0.0040,0.0402)]較A組[0.0169(0.0023,0.0426)和0.0003(0.0002,0.0043)]明顯增高(均P0.05);(5)外周血和肺部Th2細胞與纖維化評分、炎癥評分呈正相關(guān)(r值分別為0.618、0.623和0.589、0.567,均P0.05),肺部TGF-β1mRNA和IL-10mRNA與肺部纖維化評分、炎癥評分正相關(guān)(r值為0.782、0.620和0.516、0.592,均P0.05),肺部Th2細胞與肺部TGF-β1mRNA、IL-10mRNA、外周血TGF-β1、IL-10的含量正相關(guān),均P0.05。 結(jié)論皮下注射博來霉素小鼠肺部出現(xiàn)炎癥和纖維化的病理改變,Th2細胞、TGF-β1、IL-10在博來霉素致繼發(fā)性間質(zhì)性肺炎小鼠模型中的表達是增高的;Th2細胞、TGF-β1、IL-10與肺纖維化正相關(guān),提示博來霉素致繼發(fā)性間質(zhì)性肺炎的發(fā)病中存在以Th2細胞調(diào)節(jié)異常的免疫失衡。
[Abstract]:Objective to observe the changes of Th2 cells and related cytokines in murine model of interstitial pneumonia induced by bleomycin, and to explore the possible mechanism of bleomycin induced secondary interstitial pneumonia. Methods Twenty Balb/c mice were randomly divided into two groups: control group A (n = 10) and model group B (n = 10). The ratio of Th2 cells in peripheral blood and lung tissue was detected by flow cytometry, the expression of TGF- 尾 1 mRNA and IL-10mRNA in lung tissue was detected by immunofluorescence quantitative PCR assay, and the levels of LTGF- 尾 1 and IL-10 in peripheral blood were detected by Elisa. The correlation between these indexes was analyzed. Results the pulmonary fibrosis score and pulmonary inflammation score were significantly higher in group B than those in group A [0.5 and 0. 05%, respectively)] (P0. 05%). The percentage of Th2 cells in peripheral blood and lung tissue in group B [3. 32 鹵2. 1313% and 5. 72 鹵3. 37%] was significantly higher than that in group A [1. 21 鹵0. 63% and 1. 51 鹵0. 34%]. The level of TGF- 尾 1 in peripheral blood of group B [1 426.31 鹵137. 08 PG / ml] was significantly higher than that in group A [1. 21 鹵0. 63% and 1. 51 鹵0. 34%]. In group A (1138.44 鹵92.61g / ml), the level of IL-10 in peripheral blood of group B [122.09 鹵47.62 pg/ml] was significantly higher than that in group A [76.82 鹵25.74pg / ml] (all P0.05H4) and IL-10mRNAB group (0.21000.00.04184180.4700 and 0.0261n 0.00400.402) were significantly higher than those in group A [0.0169t 0.00230.0426 and 0.00030.0000.00020.0043] (all P 0.05) and pulmonary Th2 cells and fibrosis scores were significantly higher than those in group A (P 0.05). The positive correlation of inflammatory score was 0.618 ~ 0.623 and 0.589 ~ 0.567, respectively (P < 0.05). The positive correlation between lung TGF- 尾 _ 1 mRNA and IL-10mRNA and pulmonary fibrosis score was 0.7820.62 and 0.516 ~ 0.592respectively (P 0.05). The positive correlation between lung Th2 cells and lung TGF- 尾 _ 1mRNA-IL-10 mRNA and peripheral blood TGF- 尾 _ 1mRNA-IL-10 was all P 0.055.The positive correlation between TGF- 尾 _ 1mRNA and TGF- 尾 _ 1mRNA-IL-10 in lung was significant (P < 0.05), and the positive correlation between TGF- 尾 _ (1) mRNA and TGF- 尾 _ (1) IL-10 in peripheral blood was also found. Conclusion the pathological changes of pulmonary inflammation and fibrosis in bleomycin induced by subcutaneous injection of bleomycin in mice with pulmonary fibrosis may indicate that the expression of TGF- 尾 1 and IL-10 in Th2 cells is positively correlated with pulmonary fibrosis in the murine model of bleomycin induced secondary interstitial pneumonia, and the expression of TGF- 尾 1 and IL-10 is positively correlated with pulmonary fibrosis in murine models of secondary interstitial pneumonia induced by bleomycin. The results suggest that the immune imbalance of Th2 cells is abnormal in the pathogenesis of secondary interstitial pneumonia caused by bleomycin.
