膽紅素對急性肺損傷大鼠水通道蛋白1、5表達(dá)的影響

【摘要】:目的:探討膽紅素對脂多糖所致急性肺損傷大鼠的水通道蛋白1、5是否存在調(diào)節(jié)作用,探討其保護(hù)作用機(jī)制。 方法:雄性wistar大鼠18只,隨機(jī)分為生理鹽水對照組(C組),脂多糖模型組(L組),膽紅素預(yù)處理組(B組),每組各6只。每組于造模后4h,處死大鼠。測定每組肺濕/干重比值, ELISA法測血清中IL-8含量,肺組織中cAMP含量,免疫組化法測定每組水通道蛋白1、5的表達(dá)變化,觀察各組肺組織病理改變。 結(jié)果:1、血清中IL-8的含量與C組比較L組IL-8含量明顯增加,兩組間差異有統(tǒng)計學(xué)意義(P0.05)。B組IL-8含量較L組減少,但仍高于C組,其與其他兩組間比較均有統(tǒng)計學(xué)意義(P0.05)。2、肺組織中cAMP含量L組與C組比較cAMP含量明顯降低,兩組間差異有統(tǒng)計學(xué)意義(P0.05),B組cAMP含量較L組增多,但仍低于C組,其與其他兩組間比較差異均有統(tǒng)計學(xué)意義(P0.05)。3、AQP-1,AQP-5表達(dá)的變化L組與C組比較AQP-1表達(dá)明顯下降(P0.05),B組AQP-1表達(dá)明顯上調(diào),與C組之間無顯著差異(P0.05)。L組較C組AQP-5表達(dá)明顯下降(P0.05),B組AQP-5表達(dá)上升,但與C組之間差異無統(tǒng)計學(xué)意義(P0.05)。4、大鼠肺W/D值變化L組W/D比值明顯高于C組,差異有統(tǒng)計學(xué)意義(P0.05),B組較L組W/D比值降低,但仍高于C組(P0.05)。5、大鼠肺組織病理學(xué)改變C組無明顯炎癥變化,L組光鏡下可見不同程度的肺泡結(jié)構(gòu)破壞,肺泡間隔明顯增寬,肺泡腔及間質(zhì)內(nèi)大量炎性細(xì)胞浸潤,肺間質(zhì)水腫,肺泡腔內(nèi)有滲出液,局部出現(xiàn)肺不張,肺萎陷。B組肺水腫程度減輕,炎性細(xì)胞浸潤減少,肺泡腔無明顯滲出。 結(jié)論:膽紅素可能是通過調(diào)節(jié)水通道蛋白1、5表達(dá)改善水代謝,減輕肺水腫狀態(tài),,從而對急性肺損傷具有保護(hù)作用。
[Abstract]:Aim: to investigate the regulatory effect of bilirubin on aquaporin-1 5 in rats with acute lung injury induced by lipopolysaccharide (LPS) and its protective mechanism. Methods: eighteen male wistar rats were used. The rats were randomly divided into normal saline control group (n = 6), lipopolysaccharide model group (n = 6) and bilirubin pretreatment group (n = 6). The rats in each group were killed at 4 hours after modeling. The lung wet / dry weight ratio of each group was measured and the serum IL-8 content was measured by ELISA method. The content of cAMP and the expression of aquaporin 1 5 in each group were determined by immunohistochemical method. The pathological changes of lung tissue were observed in each group. Results compared with group C, the content of IL-8 in serum of group 1 was significantly higher than that of group C, and the content of IL-8 in group B was significantly lower than that in group L, but still higher than that in group C, and the content of IL-8 in group B was significantly lower than that in group C. Compared with the other two groups, the content of cAMP in lung tissue of group L and group C was significantly lower than that of group C, and the content of cAMP in group B was significantly higher than that in group L, but still lower than that in group C. There was a significant difference between the two groups in the expression of P0.05N. 3AQP-1AQP-5. The expression of AQP-1 in group L was significantly lower than that in group C. The expression of AQP-1 was up-regulated in group B (P 0.05). There was no significant difference between group C and group C in the expression of AQP-5, but there was no significant difference between group C and group C in the expression of AQP-5. The ratio of W / D in group L was significantly higher than that in group C. The ratio of W / D in group B was significantly lower than that in group L, but still higher than that in group C (P 0.05). The pathological changes of lung tissue in group C had no obvious inflammatory changes. In group L, there were different degrees of destruction of alveolar structure and widening of alveolar septum under light microscope. A large number of inflammatory cells were infiltrated in alveolar cavity and interstitial, pulmonary interstitial edema, exudate in alveolar cavity, atelectasis occurred locally, the degree of pulmonary edema in group B was reduced, the infiltration of inflammatory cells was decreased, and there was no obvious exudation in alveolar cavity. Conclusion: bilirubin may improve water metabolism and alleviate pulmonary edema by regulating the expression of aquaporin 1, which may have protective effect on acute lung injury.
