【摘要】:目的觀察氫硫酸鈉(Na HS)對脂多糖(LPS)誘導的A549肺癌細胞損傷的影響并探討其可能機制。方法 A549細胞分為對照組、LPS處理組、Na HS處理組和LPS聯(lián)合Na HS處理組。處理24 h后,采用ELISA測定培養(yǎng)液中C反應蛋白(CRP)的含量;MTT法與乳酸脫氫酶(LDH)法測定細胞損傷情況;測定A549細胞單層模型的跨上皮電阻(TER)值;免疫熒光染色法測定A549細胞中閉鎖小帶蛋白1(ZO-1)的分布情況。結(jié)果對照組與Na HS處理組比較,CRP的水平、TER值、細胞活力和ZO-1表達與分布情況無顯著性差異;與對照組相比,LPS處理組中的CRP水平明顯升高、TER值顯著減少、細胞活力下降、細胞膜ZO-1表達減少且邊界不清;與LPS處理組相比,LPS聯(lián)合Na HS處理組中的CRP水平明顯降低、TER值增加、細胞活力增加,ZO-1在細胞膜處的表達增強且分布情況部分恢復。結(jié)論 Na HS通過增強A549細胞活力、降低CRP水平、增加TER值并增加ZO-1表達抑制LPS誘導的肺損傷。
[Abstract]:Objective To observe the effect of sodium hydrogen sulfate (Na HS) on lipopolysaccharide (LPS) induced A549 damage of lung cancer cells and to explore its possible mechanism. Methods A549 cells were divided into control group, LPS treatment group, Na HS treatment group and LPS combined with Na HS group. After 24 h, determination of C reactive protein in culture using ELISA (CRP) content; MTT and lactate dehydrogenase (LDH) method for the determination of cell injury; transepithelial electrical resistance determination of A549 cell monolayer model (TER); Determination of A549 cell zonula occludens protein 1 immunofluorescence staining method (ZO-1). The distribution of the results of control group group compared with the Na HS treatment, CRP level, TER value, cell viability and ZO-1 expression had no significant difference with the distribution; compared with the control group, LPS treatment group, CRP levels were significantly increased, TER value decreased significantly, decreased cell viability, cell membrane expression of ZO-1 decreased and the boundary is not clear; compared with LPS group, LPS Lian Na HS in the CRP group were significantly decreased, TER value increased, the cell viability increased, the expression of ZO-1 in cell membrane enhanced recovery and distribution part. Conclusion Na HS enhances A549 cell viability, decrease the level of CRP, increase TER value and increase of ZO-1 lung injury induced suppression of LPS expression.
