茶黃素納米粒霧化吸入對大鼠炎性氣道黏蛋白5AC的調控

【摘要】:目的:探討高頻霧化吸入茶黃素乳酸-羥基乙酸共聚物(poly lactic-co-glycolic acid,PLGA)納米粒對炎性氣道黏蛋白5AC(mucoprotein 5AC,MUC5AC)的作用。方法:通過改良法制備茶黃素PLGA納米粒,用粒徑分析儀檢測納米粒平均粒徑,并進行包封率和載藥量測定及體外釋放試驗。將SD大鼠分為3組:對照組(A組)、香煙吸入組(B組)及干預組(C組)。RT-PCR檢測各組黏蛋白5AC m RNA含量;酶聯(lián)免疫吸附測定法(ELISA)觀察MUC5AC蛋白表達的變化,并進行組化對比大鼠氣道組織黏蛋白變化情況。結果:制備的茶黃素PLGA納米粒性質穩(wěn)定,測得其的平均粒徑為(108.24±2.86)nm,適宜霧化吸入。香煙吸入組MUC5AC的蛋白表達明顯升高,較對照組有顯著性差異(P=0.000),而給予茶黃素PLGA納米粒處理后,與香煙組比,指標明顯下調(P=0.001)。MUC5AC m RNA表達的RT-PCR電泳結果也有類似結果。氣道組織化學PAS染色也提示霧化吸入茶黃素PLGA納米粒后,氣道表面黏蛋白表達顯著下降。結論:通過改良法制備茶黃素PLGA納米粒性質穩(wěn)定,大小均勻;經(jīng)霧化途徑吸入可發(fā)揮抑制炎性氣道的黏蛋白5AC的高分泌作用,為茶黃素的開發(fā)應用提供了新的途徑。
[Abstract]:Objective: to study the high frequency atomization inhalation of theaflavin-lactic acid-glycolic acid copolymer poly lactic-co-glycolic acid. The effect of PLGA nanoparticles on inflammatory airway mucoprotein 5ACmucoprotein 5ACMUC5AC. Methods: theaflavin PLGA nanoparticles were prepared by modified method. The average particle size was measured by particle size analyzer, encapsulation efficiency, drug loading and in vitro release test were performed.
