篩選自人卵巢癌細(xì)胞系HO8910的干細(xì)胞生物學(xué)特性分析
[Abstract]:Objective to investigate the biological characteristics of human ovarian cancer cell line HO8910. Methods the stem cells of ovarian cancer cell line HO8910 were subcultured in serum-free medium and HO8910 cells were used as control. The spherical formation ability of the two kinds of cells was observed in vitro, the differentiation ability of the two kinds of cells was observed, the differentiation ability was observed, and the related markers of stem cells were detected. The sensitivity of the two kinds of cells to DDP, doxorubicin and mitoxantrone was detected by drug sensitivity test. The two kinds of cells subcultured in serum-free medium were inoculated into NOD/SCID nude mice to observe their tumorigenesis in vivo. Results the globular formation ability of HO8910 cells in serum-free medium was significantly lower than that of ovarian cancer cells (P 0.05). The positive expression rate of CD133,CD117 in ovarian cancer stem cells before differentiation was significantly higher than that after differentiation. The expression of ABCG2,Nanog,Oct4,BCRP,E-cadherin in ovarian cancer stem cells after differentiation was lower than that before differentiation, but there was no significant difference compared with that before and after differentiation of HO8910 cells. 0.25 渭 g / mL DDP, 0.5 渭 mol / L doxorubicin, 0.05 渭 g / mL mitoxantrone and 0.25 渭 g / mL mitoxantrone on ovarian cancer stem cells for 48 h. The relative activity of ovarian cancer stem cells was higher than that of HO8910 cells. With the increase of the number of stem cells inoculated with 1 脳 103 ovarian cancer stem cells in nude mice, the tumorigenesis rate increased gradually and the tumorigenesis time decreased gradually (P 0.05). Conclusion the ovarian cancer stem cells screened from human ovarian cancer cell line HO8910 have the characteristics of self-renewal and differentiation, tumorigenesis in vivo, high expression of stem cell genes and resistance to chemotherapy, which is in accordance with the theoretical standard of tumor stem cells.
