【摘要】：目的探究CD142在重度子癇前期孕婦胎盤中的表達(dá)情況及其與啟動(dòng)子區(qū)域甲基化之間的關(guān)系。方法本研究采用實(shí)時(shí)熒光定量PCR、免疫組化、蛋白質(zhì)印跡檢測(cè)24例重度子癇前期孕婦和24例正常妊娠孕婦胎盤中CD142基因的表達(dá)情況;采用甲基化特異性PCR檢測(cè)CD142啟動(dòng)子區(qū)域的甲基化情況。結(jié)果病例組胎盤中CD142mRNA相對(duì)含量(1.45±0.42)高于正常妊娠孕婦(0.25±0.28),差異有統(tǒng)計(jì)學(xué)意義(P0.05);病例組胎盤中CD142蛋白表達(dá)量(0.857±0.043)高于正常晚孕者(0.248±0.035),差異有統(tǒng)計(jì)學(xué)意義(P0.05);病例組胎盤組織中CD142啟動(dòng)子區(qū)域的甲基化陽(yáng)性率低于對(duì)照組(29.2%vs.100.0%,χ2=36.11,P0.001),非甲基化陽(yáng)性率高于正常組(100.0%vs.20.8%,χ2=29.85,P0.001);CD142的蛋白表達(dá)水平與其啟動(dòng)子區(qū)域的甲基化程度呈負(fù)相關(guān)(r=-0.909,P0.05)。結(jié)論 CD142通過(guò)甲基化調(diào)控蛋白的表達(dá)在重度子癇前期的發(fā)病過(guò)程中發(fā)揮重要作用。
[Abstract]:Objective to investigate the expression of CD142 in placenta of severe preeclampsia and its relationship with promoter region methylation. Methods the expression of CD142 gene in placenta of 24 severe preeclampsia pregnant women and 24 normal pregnant women was detected by real-time fluorescence quantitative PCR, immunohistochemistry and Western blot. Methylation of CD142 promoter region was detected by methylation specific PCR. Results the relative content of CD142mRNA in placenta of the case group (1.45 鹵0.42) was higher than that of the normal pregnant women (0.25 鹵0.28), the difference was statistically significant (P0.05). The expression of CD142 protein in placenta of case group (0.857 鹵0.043) was higher than that of normal late pregnancy (0.248 鹵0.035), the difference was statistically significant (P0.05). The positive rate of methylation in the placental tissue of the case group was lower than that in the control group (29.2vs.100.0g, 蠂 ~ 2 + 36.11p 0.001), and the positive rate of non-methylation was higher than that of the normal group (100.0vs.20.8cm, 29.85). (P0.001); The protein expression level of CD142 was negatively correlated with the methylation degree of its promoter region (r-0.909 P 0.05). Conclusion CD142 plays an important role in the pathogenesis of severe preeclampsia through the expression of methylated regulatory protein.
