子癇前期相關miR-18a對人正常滋養(yǎng)細胞浸潤功能影響的研究

【摘要】:目的通過研究子癇前期相關微小RNA-18a(micro RNA-18a,mi R-18a)對人正常滋養(yǎng)細胞(HTR8)浸潤功能的影響,探討mi R-18a與子癇前期疾病的相關性及其在發(fā)病過程中的作用機制。方法將mi R-18a前體分子分組轉染HTR8細胞,并分為三組:pre-mi RNA陰性對照組(NC)、mi R-18a過表達組和mi R-18a抑制組;反轉錄PCR檢測轉染后各組中mi R-18a m RNA的表達水平;侵襲小室實驗(Transwell)檢測轉染后各組細胞浸潤能力的變化情況。結果與NC組(0.407±0.019)比較,mi R-18a過表達組抑制效果顯著(0.884±0.061;0.161±0.014),差異具有統(tǒng)計學意義(P0.05);與NC組(66.67±3.05)比較,抑制mi R-18a的表達可明顯降低細胞的浸潤能力(29.631±3.112,P0.05)。結論 mi R-18a能夠在一定程度上影響人正常滋養(yǎng)細胞的浸潤功能,可能與子癇前期的發(fā)病相關。
[Abstract]:Objective to study the effect of preeclampsia associated microRNA-18a (micro RNA-18a,mi R-18a) on the infiltration of human normal trophoblast (HTR8). To investigate the relationship between mi R-18a and preeclampsia and its role in the pathogenesis of preeclampsia. Methods mi R-18a precursor molecules were transfected into HTR8 cells and divided into three groups: pre-mi RNA negative control group, (NC), mi R-18a overexpression group and mi R-18a inhibition group; Reverse transcription PCR was used to detect the expression level of mi R-18a m RNA in each group, and invasive chamber experiment (Transwell) was used to detect the change of cell infiltration ability after transfection. Results compared with NC group (0.407 鹵0.019), the inhibitory effect of mi R-18a overexpression group was significant (0.884 鹵0.061 鹵0.161 鹵0.014), the difference was statistically significant (P0.05). Compared with NC group (66.67 鹵3.05), inhibiting the expression of mi R-18a significantly decreased the infiltrating ability of cells (29.631 鹵3.112 P 0.05). Conclusion mi R-18a can affect the infiltration function of human normal trophoblast to some extent and may be related to the pathogenesis of preeclampsia.
