【摘要】：目的探討siRNA沉默結(jié)腸癌轉(zhuǎn)移相關(guān)基因1(MACC1)表達(dá)對(duì)Hela宮頸癌細(xì)胞增殖及侵襲的影響。方法通過脂質(zhì)體將siRNA MACC1及siRNA NC轉(zhuǎn)入Hela細(xì)胞中,RT-PCR及western blot檢測(cè)轉(zhuǎn)染效果,MTT法檢測(cè)細(xì)胞活力,流式細(xì)胞術(shù)檢測(cè)細(xì)胞周期,Transwell法檢測(cè)細(xì)胞侵襲能力,western blot檢測(cè)細(xì)胞周期蛋白D1(cyclin D1)、cyclin E,基質(zhì)金屬蛋白酶2(MMP2)、MMP9蛋白表達(dá)及細(xì)胞外調(diào)節(jié)蛋白激酶(ERK)磷酸化水平。結(jié)果與siRNA NC組比較,siRNA MACC1組中MACC1蛋白及mRNA表達(dá)量降低(P0.01),細(xì)胞活力及侵襲能力下降(P0.01),細(xì)胞周期阻滯在G1期,cyclin D1、cyclin E、MMP2、MMP9及p-ERK表達(dá)量下調(diào)(P0.01)。結(jié)論siRNA MACC1能顯著的抑制宮頸癌Hela細(xì)胞增殖與侵襲,可能與下調(diào)ERK磷酸化水平有關(guān)。
[Abstract]:Objective to investigate the effect of siRNA silencing the expression of metastasis associated gene 1 (MACC1) on the proliferation and invasion of Hela cervical cancer cells. Methods siRNA MACC1 and siRNA NC were transferred into Hela cells by liposome. Transfection effect was detected by RT-PCR and western blot. Cell viability was detected by MTT assay, cell cycle was detected by flow cytometry, and cell invasion ability was detected by Transwell assay. The expression of cyclin D1), cyclin E, matrix metalloproteinase 2 (MMP2), the expression of MMP9 protein and the phosphorylation of extracellular regulated protein kinase (ERK) were detected by western blot. Results compared with siRNA NC group, the expression of MACC1 protein and mRNA in siRNA MACC1 group was decreased (P0.01), cell viability and invasiveness were decreased (P0.01), cell cycle was blocked in G1 phase, and cyclin D1 cyclin EmMP2 was found in siRNA MACC1 group. The expression of MMP9 and p-ERK was down-regulated (P0.01). Conclusion siRNA MACC1 can significantly inhibit the proliferation and invasion of cervical cancer Hela cells, which may be related to the down-regulation of ERK phosphorylation.
