孕酮對(duì)人臍靜脈收縮的抑制效應(yīng)及其機(jī)制研究

【摘要】:【目的】探討孕酮(Pro,progesterone)對(duì)人臍靜脈收縮功能的影響及其相關(guān)機(jī)制!痉椒ā渴占R床足月兒臍帶(N=106),分離臍靜脈,將分離后的血管剪成3-5mm長(zhǎng)度的血管環(huán),置于37℃5ml Krebs溶液恒溫水浴槽內(nèi)進(jìn)行血管功能實(shí)驗(yàn),研究孕酮對(duì)去氧腎上腺素(PE,phenylephrine)、五羥色胺(5-HT,serotonin)誘導(dǎo)的人臍靜脈收縮的作用及機(jī)制。同時(shí),本實(shí)驗(yàn)采取了RT-PCR及Western-blot技術(shù)檢測(cè)了孕酮對(duì)人臍靜脈中L-Ca2+通道(CaV1.2)及孕酮受體(PR,progesterone receptor)的表達(dá)水平的影響!窘Y(jié)果】孕酮顯著抑制PE及5-HT誘導(dǎo)的人臍靜脈收縮,這種抑制作用不能被內(nèi)皮一氧化氮(nitric oxide,NO)及前列環(huán)素(prostacyclin,PGI2)合成的阻斷劑、血管內(nèi)皮的去除及孕激素受體拮抗劑或鉀通道阻斷劑所拮抗。同時(shí),在Krebs溶液中,對(duì)PE、5-HT、Bay-k8644(Pseudoginsenoside,氟硝尼啶,電壓依賴型鈣離子通道的激動(dòng)劑)、KCl誘導(dǎo)的臍靜脈收縮的抑制作用,以及在無(wú)鈣Krebs溶中,對(duì)CaCl2誘導(dǎo)的臍靜脈收縮的抑制作用,孕酮與尼非地平(nife,nifedipine,特異性CaV1.2阻斷劑)或孕酮聯(lián)合尼非地平的作用無(wú)差異。但是,mibe(mibefradil,米貝地爾,L-Ca2+通道(CaV1.2)和T-Ca2+通道(CaV3.2)阻斷劑)對(duì)PE及5-HT誘導(dǎo)的臍靜脈收縮的抑制作用顯著強(qiáng)于孕酮或(和)尼非地平。而且,在無(wú)鈣Krebs溶液中,孕酮對(duì)PE、5-HT及Caffine(咖啡因)誘導(dǎo)的人臍靜脈收縮無(wú)抑制作用。同時(shí),臍靜脈孵育60分鐘孕酮不改變CaV1.2及PR的mRNA和蛋白表達(dá)水平。【結(jié)論】1)孕酮對(duì)人臍靜脈的收縮有抑制作用,這種抑制作用主要通過(guò)阻斷L-Ca2+通道,而不是T-Ca2+通道,從而抑制外鈣內(nèi)流,同時(shí),孕酮不是通過(guò)促進(jìn)血管內(nèi)皮性舒張、與孕酮受體相結(jié)合、激活鉀通道和抑制內(nèi)鈣釋放來(lái)起作用。
