Th22細(xì)胞通過(guò)釋放IL-22活化宮頸癌細(xì)胞AKT通路促進(jìn)其增殖

【摘要】:目的研究IL-22及Th22細(xì)胞在宮頸癌組織中的表達(dá)和頻率,分析其與宮頸癌臨床分期的相關(guān)性,探討它們與宮頸癌發(fā)生發(fā)展的關(guān)系。方法以西南醫(yī)院2016年1月至2月收集的32例宮頸癌患者為研究對(duì)象,免疫組化、Western blot檢測(cè)IL-22在宮頸癌組織、癌旁組織和正常宮頸組織中的表達(dá)情況;流式細(xì)胞技術(shù)檢測(cè)Th22細(xì)胞在宮頸癌患者外周血CD4~+T淋巴細(xì)胞和腫瘤浸潤(rùn)淋巴細(xì)胞中的頻率及其表達(dá)IL-22情況,探討其與宮頸癌臨床分期的關(guān)系;細(xì)胞共培養(yǎng)探究IL-22是否通過(guò)促進(jìn)AKT信號(hào)通路活化而促進(jìn)宮頸癌細(xì)胞增殖。結(jié)果宮頸癌腫瘤組織中IL-22的表達(dá)水平顯著高于癌旁和正常宮頸組織,而且Ⅲ~Ⅳ期宮頸癌組織IL-22的表達(dá)明顯高于Ⅰ~Ⅱ期。宮頸癌腫瘤組織浸潤(rùn)淋巴細(xì)胞中,Th22細(xì)胞頻率顯著高于宮頸癌患者及正常人的外周血,且與宮頸癌的臨床分期正相關(guān)。Th22是宮頸癌組織中IL-22主要來(lái)源。細(xì)胞共培養(yǎng)實(shí)驗(yàn)顯示IL-22促進(jìn)了AKT的磷酸化并誘導(dǎo)下游分子的表達(dá)。結(jié)論 Th22細(xì)胞通過(guò)釋放IL-22活化宮頸癌細(xì)胞內(nèi)AKT通路促使其增殖,干預(yù)Th22及其效應(yīng)分子IL-22是宮頸癌防治的一個(gè)潛在靶標(biāo)。
[Abstract]:Objective to study the expression and frequency of IL-22 and Th22 cells in cervical carcinoma, and to analyze their correlation with the clinical stage of cervical cancer, and to explore the relationship between them and the occurrence and development of cervical cancer. Methods 32 patients with cervical cancer collected from January to February 2016 in Southwest Hospital were studied.
