青海地區(qū)不同海拔藏漢民族子癇前期患者人類白細(xì)胞抗原G表達(dá)研究

【摘要】:目的探討青海地區(qū)不同海拔藏漢民族子癇前期患者的人類白細(xì)胞抗原G(HLA-G)表達(dá)情況。方法選取2015年1月—2016年1月在青海大學(xué)附屬醫(yī)院分娩的漢族產(chǎn)婦30例(海拔2 200 m),其中子癇前期16例,正常妊娠14例;另選取同期在玉樹(shù)藏族自治州人民醫(yī)院分娩的藏族產(chǎn)婦21例(海拔3 700 m),其中子癇前期10例,正常妊娠11例。采用實(shí)時(shí)定量聚合酶鏈?zhǔn)椒磻?yīng)法和蛋白質(zhì)印跡法檢測(cè)不同情況產(chǎn)婦胎盤(pán)組織中的HLA-G mRNA和蛋白表達(dá)。結(jié)果不同情況產(chǎn)婦HLA-G mRNA和蛋白表達(dá)比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。低海拔漢族子癇前期、高海拔藏族正常妊娠、高海拔藏族子癇前期產(chǎn)婦HLA-G mRNA和蛋白表達(dá)低于低海拔漢族正常妊娠產(chǎn)婦,差異有統(tǒng)計(jì)學(xué)意義(P0.05);高海拔藏族子癇前期產(chǎn)婦HLA-G mRNA和蛋白表達(dá)低于高海拔藏族正常妊娠產(chǎn)婦,差異有統(tǒng)計(jì)學(xué)意義(P0.05);高海拔藏族子癇前期產(chǎn)婦HLA-G蛋白表達(dá)低于低海拔漢族子癇前期產(chǎn)婦,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。而低海拔漢族子癇前期產(chǎn)婦與高海拔藏族正常妊娠產(chǎn)婦HLA-G mRNA和蛋白表達(dá)比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);低海拔漢族子癇前期產(chǎn)婦與高海拔藏族子癇前期產(chǎn)婦HLA-G mRNA表達(dá)比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論高原慢性低氧環(huán)境可能影響HLA-G基因的表達(dá),不同海拔藏漢民族HLA-G的差異性表達(dá)可能在青海高原地區(qū)子癇前期的發(fā)病過(guò)程中起重要作用,是導(dǎo)致高原地區(qū)子癇前期發(fā)病率高的原因之一。
[Abstract]:Objective to investigate the expression of human leukocyte antigen G (HLA-G) in preeclampsia patients of Tibetan and Han nationalities at different altitudes in Qinghai. Methods from January 2015 to January 2016, 30 pregnant women of Han nationality (2 200 m), above sea level) who gave birth in the affiliated hospital of Qinghai University were selected, including 16 cases of preeclampsia and 14 cases of normal pregnancy. In addition, 21 pregnant women (3 700 m), above sea level) who were delivered in Yushu Tibetan Autonomous Prefecture people's Hospital at the same time were selected, including 10 cases of preeclampsia and 11 cases of normal pregnancy. The expression of HLA-G mRNA and protein in placenta tissues of puerpera was detected by real-time quantitative polymerase chain reaction and Western blotting. Results the expression of HLA-G mRNA and protein in parturient was significantly different (P 0.05). The expression of HLA-G mRNA and protein in preeclampsia of Han nationality at low altitude was lower than that in normal pregnant women of Han nationality at low altitude. The expression of HLA-G mRNA and protein in Tibetan preeclampsia at high altitude was lower than that in normal pregnant women at high altitude. The expression of HLA-G protein in Tibetan preeclampsia at high altitude was significantly lower than that in preeclampsia of Han nationality at low altitude (P0.05). There was no significant difference in HLA-G mRNA and protein expression between preeclampsia women of Han nationality at low altitude and normal pregnant women with Tibetan nationality at high altitude (P0.05), and HLA-G mRNA expression in preeclampsia women of Han nationality at low altitude and Tibetan preeclampsia at high altitude. The difference was not statistically significant (P0.05). Conclusion chronic hypoxia at high altitude may affect the expression of HLA-G gene, and the differential expression of HLA-G may play an important role in the pathogenesis of preeclampsia in Qinghai Plateau. It is one of the causes of high incidence of preeclampsia in plateau area.
