GRIM-19蛋白在taurolidine誘導(dǎo)宮頸癌細(xì)胞凋亡中的作用

【摘要】:目的探討GRIM-19蛋白在taurolidine誘導(dǎo)人宮頸癌Hela細(xì)胞凋亡中的作用。方法應(yīng)用四甲基偶氮唑藍(lán)(MTT)法檢測(cè)不同劑量組宮頸癌細(xì)胞(Hela細(xì)胞系)存活率的影響;采用流式細(xì)胞術(shù)檢測(cè)不同劑量組Hela細(xì)胞的凋亡數(shù)據(jù);采用Western印跡法檢測(cè)各實(shí)驗(yàn)組中Hela細(xì)胞中GRIM-19蛋白的表達(dá)變化;同時(shí)采用酶聯(lián)免疫吸附(ELISA)法檢測(cè)半胱氨酸蛋白酶(caspase)9的活性。結(jié)果 MTT結(jié)果顯示,taurolidine對(duì)Hela細(xì)胞生長(zhǎng)有顯著的抑制作用,流式細(xì)胞術(shù)結(jié)果顯示出實(shí)驗(yàn)組Hela細(xì)胞凋亡比例顯著上升,并呈現(xiàn)出明顯的時(shí)程和量效關(guān)系(P0.05)。Western印跡分析顯示,與對(duì)照組相比,GRIM-19蛋白在各實(shí)驗(yàn)組Hela細(xì)胞中的表達(dá)顯著提高(P0.05),同樣顯示出了一定的時(shí)程和量效關(guān)系。ELISA檢測(cè)結(jié)果顯示,caspase-9在各實(shí)驗(yàn)組Hela細(xì)胞中的表達(dá)顯著增加,與GRIM-19蛋白的表達(dá)顯示出了相當(dāng)?shù)囊恢滦。結(jié)論 Taurolidine可通過(guò)促進(jìn)GRIM-19蛋白的表達(dá)誘導(dǎo)Hela細(xì)胞的凋亡,且線粒體凋亡很可能是taurolidine誘導(dǎo)Hela細(xì)胞凋亡的重要途徑。
[Abstract]:Objective to investigate the role of GRIM-19 protein in apoptosis of human cervical carcinoma Hela cells induced by taurolidine. Methods MTT assay was used to detect the survival rate of cervical cancer cell line (Hela cell line) in different dose groups, and flow cytometry was used to detect the apoptosis data of Hela cell line. The expression of GRIM-19 protein and the activity of cysteine protease (caspase) 9 were detected by Western blot and enzyme-linked immunosorbent assay (Elisa). Results MTT results showed that taurolidine inhibited Hela cell growth significantly, and flow cytometry showed that the proportion of Hela cell apoptosis in the experimental group increased significantly, and showed a significant time-course and dose-effect relationship (P0.05). Western blot analysis showed that Hela cells in the experimental group had significant apoptosis. Compared with the control group, the expression of pGRIM-19 protein in Hela cells was significantly increased (P0.05). The results of Elisa showed that the expression of pGRIM-19 in Hela cells of each experimental group was significantly higher than that of the control group (P0.05), and also showed a certain time-course and dose-response relationship. The results of Elisa showed that the expression of pGRIM-19 in Hela cells increased significantly. The expression of GRIM-19 protein was consistent with that of GRIM-19 protein. Conclusion Taurolidine can induce the apoptosis of taurolidine cells by promoting the expression of GRIM-19 protein, and mitochondrial apoptosis may be an important way to induce apoptosis of Hela cells.
