【摘要】：目的： 通過檢測CIP2A與c-Myc在子宮內(nèi)膜樣腺癌中的表達(dá)情況，，分析其與臨床病理參數(shù)的關(guān)系，并初步探討兩因子在子宮內(nèi)膜樣腺癌中發(fā)生發(fā)展的作用。 方法： 采用實(shí)時(shí)定量逆轉(zhuǎn)錄聚合酶反應(yīng)方法檢測80例子宮內(nèi)膜樣腺癌和43例正常子宮內(nèi)膜中的CIP2A mRNA的表達(dá)情況，并比較它們的差異。采用免疫組織化學(xué)方法檢測55例子宮內(nèi)膜樣腺癌（Endometrial adenocarcinoma，EAC），20例子宮內(nèi)膜不典型增生（Endometrial atypical hyperplasia，EAH）和24例正常子宮內(nèi)膜（Normal endometrium，NE）中CIP2A及c-Myc蛋白的表達(dá)情況，比較它們的差異；并分析CIP2A與c-Myc在子宮內(nèi)膜樣腺癌中的表達(dá)與臨床病理參數(shù)的關(guān)系及兩者的相關(guān)性。所有數(shù)據(jù)均通過SPSS17.0統(tǒng)計(jì)軟件進(jìn)行t檢驗(yàn)、秩和檢驗(yàn)及Spearman等級(jí)相關(guān)分析，以P0.05表示差異有統(tǒng)計(jì)學(xué)意義。 結(jié)果： 1. CIP2AmRNA在子宮內(nèi)膜樣腺癌中相對(duì)表達(dá)水平明顯高于正常子宮內(nèi)膜組織，差異具有統(tǒng)計(jì)學(xué)意義（P=0.016）。 2. CIP2A及c-Myc蛋白在NE、EAH、EAC組織中表達(dá)強(qiáng)度呈上升趨勢，差異具有統(tǒng)計(jì)學(xué)意義（P0.05）；在EAC組織中，CIP2A、c-Myc的表達(dá)強(qiáng)度均與肌層浸潤深度、臨床分期及組織分化程度有關(guān)（P0.05），而與淋巴結(jié)有無轉(zhuǎn)移、年齡無明顯相關(guān)性(P0.05)； 3. Spearman等級(jí)相關(guān)分析顯示在EAC組織中CIP2A與c-Myc的表達(dá)強(qiáng)度呈正相關(guān)（r=0.607，P0.0001）。 結(jié)論：CIP2A與c-Myc在子宮內(nèi)膜樣腺癌中過表達(dá)，兩者呈正相關(guān)；CIP2A可能通過穩(wěn)定c-Myc蛋白參與子宮內(nèi)膜癌的發(fā)生和發(fā)展。
[Abstract]:Objective: to detect the expression of CIP2A and c-Myc in endometrial adenocarcinoma and to analyze the relationship between CIP2A and clinicopathologic parameters, and to explore the role of two factors in the occurrence and development of endometrial adenocarcinoma. Methods: the expression of CIP2A mRNA in 80 cases of endometrial adenocarcinoma and 43 cases of normal endometrium was detected by real-time quantitative reverse transcriptase polymerase reaction. The expression of CIP2A and c-Myc protein in 55 cases of Endometrial adenocarcinoma (EAC) and 20 cases of Endometrial atypical hyperplasia EAH (EAH) and 24 cases of normal endometrium (NE) were detected by immunohistochemical method. The relationship between the expression of CIP2A and c-Myc in endometrioid adenocarcinoma and clinicopathological parameters was analyzed. All the data were analyzed by SPSS 17.0 statistical software for t test, rank sum test and Spearman rank correlation analysis. Results: 1. The relative expression of CIP2 mRNA in endometrial adenocarcinoma was significantly higher than that in normal endometrial tissue (P0. 016). 2. The expression intensity of CIP2A and c-Myc protein increased in EAH EAC tissue, the difference was statistically significant (P0.05), the expression intensity of CIP2A and c-Myc protein were related to the depth of myometrium invasion, clinical stage and tissue differentiation (P0.05), but correlated with lymph node metastasis, and the expression intensity of CIP2A and c-Myc protein in EAC tissue was correlated with the depth of myometrial invasion, the clinical stage and the degree of tissue differentiation (P0.05). Age had no significant correlation (P0.05); 3. Spearman grade correlation analysis showed that there was a positive correlation between the expression of CIP2A and c-Myc in EAC tissues (r = 0.607, P 0.0001). Conclusion the overexpression of CIP2A and c-Myc in endometrial adenocarcinoma is positively correlated with the formation and development of endometrial carcinoma by stabilizing c-Myc protein.
