葡萄糖轉(zhuǎn)運(yùn)蛋白2和蛋白酪氨酸磷酸酶-1B在甲狀腺毒癥妊娠大鼠胰腺中表達(dá)的觀察

【摘要】:目的檢測(cè)甲狀腺毒癥妊娠大鼠胰腺中葡萄糖轉(zhuǎn)運(yùn)蛋白2(GLUT2)和蛋白酪氨酸磷酸酶-1B(PTP-1B)的表達(dá),探討其影響糖代謝的可能機(jī)制。方法雌性SD大鼠分為非妊娠組和待妊娠組,待妊娠組按雌雄比例2∶1合籠受孕后進(jìn)一步分為單純?nèi)焉?R)組、妊娠期左甲狀腺素鈉低劑量(RA)組、妊娠期左甲狀腺素鈉中劑量(RB)組和妊娠期左甲狀腺素鈉高劑量(RC)組,非妊娠組進(jìn)一步分為正常對(duì)照(N)組、左甲狀腺素鈉低劑量(A)組、左甲狀腺素鈉中劑量(B)組和左甲狀腺素鈉高劑量(C)組。予A、RA組(左甲狀腺素鈉50μg/100g)、B、RB組(左甲狀腺素鈉100μg/100g)、C、RC組(左甲狀腺素鈉150μg/100g)灌胃制造甲狀腺毒癥模型,N、R組予等量生理鹽水。檢測(cè)各組雌鼠甲狀腺功能、FPG和FIns,Western blot檢測(cè)胰腺中GLUT2和PTP-1B的表達(dá)。結(jié)果隨著左甲狀腺素鈉劑量增加,N、A、B、C組和R、RA、RB、RC組游離三碘甲狀腺原氨酸(FT3)[(2.65±0.07)vs(4.05±0.10)vs(4.87±0.63)vs(5.62±0.73)pmol/L;(2.67±0.10)vs(4.07±0.09)vs(4.84±0.11)vs(5.65±0.18)pmol/L,P0.01]、游離甲狀腺素(FT4)[(10.54±0.58)vs(13.46±0.78)vs(25.77±0.85)vs(36.58±0.91)pmol/L;(10.61±0.79)vs(13.53±0.66)vs(25.90±1.12)vs(36.65±0.62)pmol/L,P0.01]升高,促甲狀腺激素(TSH)[(0.086±0.009)vs(0.029±0.002)vs(0.024±0.001)vs(0.018±0.002)μU/ml;(0.070±0.009)vs(0.027±0.002)vs(0.021±0.001)vs(0.015±0.002)μU/ml,P0.01]降低。甲狀腺毒癥各組(A、B、C組和RA、RB、RC組)FPG較對(duì)照組(N組和R組)升高[(5.43±0.32)、(5.74±0.21)、(6.34±0.46)vs(4.98±0.25)mmol/L;(5.37±0.48)、(5.78±0.39)、(6.45±0.44)vs(4.88±0.38)mmol/L,P0.01],C組FIns較N組升高[(10.40±1.52)vs(8.25±0.80)μU/ml,P0.01],妊娠各組(R、RA、RB、RC組)FIns較非妊娠各組(N、A、B、C組)升高[(9.16±1.28)vs(8.25±0.80)μU/ml;(10.40±1.31)vs(9.20±1.32)μU/ml;(10.41±1.43)vs(9.75±1.24)μU/ml;(11.09±1.17)vs(10.40±1.52)μU/ml,P0.01]。Western blot結(jié)果顯示,甲狀腺毒癥各組GLUT2表達(dá)較N、R組降低,PTP-1B表達(dá)升高(P0.05)。結(jié)論 GLUT2及PTP-1B可能與妊娠期合并甲狀腺毒癥雌鼠出現(xiàn)糖代謝異常相關(guān)。
[Abstract]:Objective to detect the expression of glucose transporter 2 (GLUT2) and protein tyrosine phosphatase 1B (PTP-1B) in the pancreas of pregnant rats with thyroid toxicity, and to explore the possible mechanism of the effects of GLUT2 and PTP-1B on glucose metabolism. Methods female Sprague-Dawley rats were divided into two groups: the non-pregnant group and the unpregnant group. The female SD rats were further divided into simple pregnancy (R) group and low dose of levothyroxine sodium (RA) group according to the sex ratio of 2:1. The middle dose of levothyroxine sodium (RB) during pregnancy and the high dose of levothyroxine sodium during pregnancy (RC) group were further divided into normal control (N) group and low-dose (A) group. Middle dose (B) group and high dose (C) group of levothyroxine sodium. The model of thyrotoxicosis was made by oral administration of Thyroxine sodium (50 渭 g / 100 g) in Acara group (50 渭 g / 100 g left thyroxine sodium) and 100 渭 g / 100 g left thyroxine sodium group (100 渭 g / 100 g left thyroxine sodium group) and the same amount of normal saline was given to the control group (150 渭 g / 100g levathyroxine sodium).
