Beclin-1、Bcl-2、Bax在子宮內(nèi)膜樣腺癌中的表達(dá)及意義

【摘要】:【目的】 1.探討B(tài)eclin-1、Bcl-2、Bax三種基因蛋白在子宮內(nèi)膜樣腺癌中的表達(dá)及意義。 2.探討子宮內(nèi)膜樣腺癌中Beclin-1、Bcl-2、Bax三種基因蛋白的表達(dá)水平的差異,,三者之間的相關(guān)性,以及與臨床病理特征之間的關(guān)系。 【方法】 運(yùn)用免疫組化S-P法檢測(cè)80例子宮內(nèi)膜樣腺癌及40例正常子宮內(nèi)膜組織制作成的組織芯片中Beclin-1、Bcl-2、Bax三種基因蛋白的表達(dá),并分析三者間的相關(guān)性,以及與臨床病理特征之間的關(guān)系。 【結(jié)果】 1、在子宮內(nèi)膜樣腺癌和正常子宮內(nèi)膜組織中Beclin-1的陽(yáng)性表達(dá)率分別為7.5%和95%;Bcl-2為88.75%和27.5%;Bax為82.5%,20%。Beclin-1在子宮內(nèi)膜樣腺癌以及正常子宮內(nèi)膜組織中的表達(dá)率差異有統(tǒng)計(jì)學(xué)意義(P0.05)。Bcl-2的表達(dá)在子宮內(nèi)膜樣腺癌以及正常子宮內(nèi)膜組織中的表達(dá)率差異有統(tǒng)計(jì)學(xué)意義(P0.05)。Bax在子宮內(nèi)膜樣腺癌以及正常子宮內(nèi)膜組織中的表達(dá)率差異有統(tǒng)計(jì)學(xué)意義(P0.05)。 2、本實(shí)驗(yàn)子宮內(nèi)膜樣腺癌Bax的表達(dá)與Bcl-2的表達(dá)無(wú)明顯相關(guān)性(P0.05),Beclin-1與Bcl-2的表達(dá)顯著負(fù)相關(guān)(P0.05),Beclin-1與Bax的表達(dá)顯著負(fù)相關(guān)(P0.05)。 3、80例子宮內(nèi)膜樣腺癌中Beclin-1的陽(yáng)性表達(dá)率與腫瘤分化程度及TNM分期密切相關(guān)(P0.05),與患者年齡、腫瘤大小、浸潤(rùn)深度及淋巴結(jié)是否轉(zhuǎn)移均無(wú)相關(guān)性(P0.05)。Bcl-2的陽(yáng)性表達(dá)率與腫瘤分化程度密切相關(guān)(P0.05),組間差異有統(tǒng)計(jì)學(xué)意義(P0.05),與患者年齡、腫瘤大小、浸潤(rùn)深度、淋巴結(jié)是否轉(zhuǎn)移及腫瘤TNM分期均無(wú)相關(guān)性(P0.05)。Bax的陽(yáng)性表達(dá)率與患者年齡、腫瘤大小、浸潤(rùn)深度、淋巴結(jié)是否轉(zhuǎn)移、腫瘤分化程度及TNM分期均無(wú)相關(guān)性(P0.05)。 【結(jié)論】 1、Bax, Bcl-2在子宮內(nèi)膜樣腺癌中高表達(dá),而B(niǎo)eclin-1在子宮內(nèi)膜樣腺癌中低表達(dá),提示三者可能均參與了子宮內(nèi)膜樣腺癌的發(fā)生發(fā)展。 2、Beclin-1與Bcl-2、Beclin-1與Bax表達(dá)呈負(fù)相關(guān),提示Bax、Bcl-2可能通過(guò)抑制Beclin-1的表達(dá),共同促進(jìn)子宮內(nèi)膜樣腺癌的發(fā)生發(fā)展。 3、Beclin-1與Bcl-2的表達(dá)與子宮內(nèi)膜樣腺癌的某些臨床病理特征之間存在一定相關(guān)性。Beclin-1陽(yáng)性表達(dá)與腫瘤分化程度及TNM分期密切相關(guān),可望作為預(yù)后評(píng)估的參考指標(biāo)。
[Abstract]:[objective] 1. To investigate the expression and significance of Beclin-1 Bcl-2mBax gene protein in endometrial adenocarcinoma. 2. To investigate the difference of the expression level of three kinds of gene proteins of Beclin-1, Bcl-2 and Bax in endometrial adenocarcinoma, and the correlation between them. Immunohistochemical S-P method was used to detect three kinds of Beclin-1 Bcl-2Bcl-2BX in 80 cases of endometrial adenocarcinoma and 40 cases of normal endometrial tissue. Gene protein expression, The correlation between them and clinicopathological features was analyzed. [results] 1. The positive expression rates of Beclin-1 in endometrioid adenocarcinoma and normal endometrium were 7.5% and 95%, respectively. Bcl-2 was 88.75% and 27.5% respectively. There was significant difference in the expression of Bax in endometrial adenocarcinoma and normal endometrium (P0.05) .Bcl-2 expression in endometrial adenocarcinoma and normal endometrium (P0.05). There was significant difference in the expression rate of Beclin-1 in endometrial adenocarcinoma and normal endometrium (P0.05). The expression rate of Bax in endometrial adenocarcinoma and normal endometrium was significantly different (P0.05). There was no significant correlation between the expression of Bax and Bcl-2 in endometrial adenocarcinoma (P0.05). The expression of Beclin-1 was negatively correlated with the expression of Bax (P0.05). The positive expression rate of Beclin-1 was closely related to tumor differentiation and TNM stage in 80 patients with endometrial adenocarcinoma (P0.05), and correlated with the age of the patients. There was no correlation between tumor size, depth of invasion and lymph node metastasis (P0.05). The positive expression rate of Bcl-2 was closely related to the degree of tumor differentiation (P0.05). The difference between groups was statistically significant (P0.05). There was no correlation between lymph node metastasis and TNM stage (P0.05). The positive expression rate of Bax was correlated with age, tumor size, depth of invasion, lymph node metastasis or not. There was no correlation between tumor differentiation and TNM staging (P0.05). [conclusion] 1Baxand Bcl-2 were highly expressed in endometrial adenocarcinoma, but Beclin-1 was low expression in endometrioid adenocarcinoma. These results suggest that all three may be involved in the development of endometrial adenocarcinoma. 2Beclin-1 and Bcl-2P Beclin-1 are negatively correlated with Bax expression, suggesting that BaxanBcl-2 may inhibit the expression of Beclin-1 by inhibiting the expression of Beclin-1. The expression of Beclin-1 and Bcl-2 was correlated with some clinicopathological features of endometrial adenocarcinoma. The positive expression of Beclin-1 was closely related to the degree of tumor differentiation and TNM stage. It is expected to be a reference index for prognostic evaluation.
