【摘要】：目的卵巢癌是婦科腫瘤中死亡率居首的常見惡性腫瘤,尋找新的生物學(xué)標(biāo)志物以提高卵巢癌診治的敏感性與特異性至關(guān)重要。本研究探討GTP酶激活蛋白SH3功能區(qū)結(jié)合蛋白(Ras-GTPase-activating protein SH3domain binding protein,G3BP)和骨橋蛋白(osteopontin,OPN)在上皮性卵巢組織中的表達(dá)、意義及其表達(dá)的相關(guān)性,為卵巢腫瘤的臨床診斷及治療提供依據(jù)。方法采用免疫組織化學(xué)方法檢測(cè)G3BP和OPN在來源于中國(guó)醫(yī)科大學(xué)盛京醫(yī)院2009-10-01-2015-05-31的11例正常卵巢組織、20例卵巢上皮性良性腫瘤組織、31例卵巢上皮性交界性腫瘤組織及60例卵巢上皮性惡性腫瘤組織中的表達(dá)情況;并對(duì)二者表達(dá)的相關(guān)性進(jìn)行統(tǒng)計(jì)分析。結(jié)果 G3BP在正常卵巢上皮組織、卵巢上皮性良性腫瘤組織、卵巢上皮性交界性腫瘤組織及卵巢上皮性惡性腫瘤組織中的表達(dá)呈遞增趨勢(shì),其中上皮性良性腫瘤組織與正常卵巢上皮組織比較,χ~2=66.38,P0.01;上皮性交界性腫瘤組織與上皮性良性腫瘤組織比較,χ~2=41.183,P0.01;上皮性惡性腫瘤組織與上皮性交界性腫瘤組織比較,χ~2=73.342,P0.01。OPN在正常卵巢上皮組織、卵巢上皮性良性腫瘤組織、卵巢上皮性交界性腫瘤組織及卵巢上皮性惡性腫瘤組織中的表達(dá)呈遞增趨勢(shì),其中上皮性良性腫瘤組織與正常卵巢上皮組織比較,χ~2=60.005,P0.01;上皮性交界性腫瘤組織與上皮性良性腫瘤組織比較,χ~2=43.844,P0.01;上皮性惡性腫瘤組織與上皮性交界性腫瘤組織比較,χ~2=73.429,P0.01。G3BP與OPN在卵巢上皮性惡性腫瘤組織中的表達(dá)具有正相關(guān)性,r=0.351,P0.01。結(jié)論 G3BP和OPN表達(dá)升高與上皮性卵巢癌的發(fā)生發(fā)展有密切關(guān)系,且二者可能存在協(xié)同作用。
[Abstract]:Objective Ovarian cancer is the most common malignant tumor in gynecological tumors. It is very important to search for new biomarkers to improve the sensitivity and specificity of diagnosis and treatment of ovarian cancer. The purpose of this study was to investigate the expression of GTPase-activating protein SH3domain binding protein G3BP and osteopontin binding protein in epithelial ovarian tissues, and to provide evidence for clinical diagnosis and treatment of ovarian tumors. Methods Immunohistochemical method was used to detect G3BP and OPN in 11 normal ovarian tissues from Shengjing Hospital of China Medical University 2009-10-01-2015-05-31. 20 cases of ovarian epithelial benign tumor tissues and 31 cases of ovarian epithelial sexual intercourse borderline tumor tissues and 60 cases of ovarian epithelial sexual intercourse tissues were detected. Cases of ovarian epithelial malignant tumors expressed in the tissues; The correlation between them was analyzed statistically. Results the expression of G3BP in normal ovarian epithelium, ovarian epithelial benign tumor, ovarian epithelial sexual intercourse borderline tumor and ovarian epithelial malignant tumor showed an increasing tendency. The comparison between epithelial benign tumor and normal ovarian epithelial neoplasm was 66.38 (P0.01); that of epithelial sexual intercourse between epithelial benign tumor and epithelial benign tumor was 41.183 (P0.01); and that of epithelial malignant tumor was significantly higher than that of epithelial sexual intercourse (蠂 ~ 23.342P0.01.OPN). Normal ovarian epithelium, The expression of ovarian epithelial benign tumor, borderline ovarian epithelial tumor and ovarian epithelial malignant tumor showed an increasing trend. The comparison between epithelial benign tumor tissue and normal ovarian epithelial tissue, 蠂 ~ (2 +) ~ (22) (60.005) P _ (0.01), epithelial sexual intercourse boundary tumor tissue and epithelial benign tumor tissue, 蠂 ~ (2 +) 43.844 (P ~ (0.01), and epithelial malignant tumor tissue and epithelial sexual intercourse boundary tumor tissue, 蠂 ~ (2 +) ~ (22) 429 (P 0.01.G3BP), compared with epithelial sexual intercourse tissue (P _ (0.01), compared with epithelial benign tumor tissue and epithelial benign tumor tissue (P _ (0.01). The expression of OPN in epithelial ovarian neoplasms was positively correlated with that of 0.351P0.01. Conclusion the increased expression of G3BP and OPN is closely related to the occurrence and development of epithelial ovarian cancer, and there may be a synergistic effect between the expression of G3BP and OPN.
