內(nèi)質(zhì)網(wǎng)應(yīng)激對(duì)碳離子輻照宮頸癌HeLa細(xì)胞的影響

【摘要】:利用不同劑量的碳離子輻照二硫蘇糖醇(2.5 mmol/L)預(yù)處理的HeLa細(xì)胞,探討了內(nèi)質(zhì)網(wǎng)應(yīng)激反應(yīng)對(duì)碳離子輻照宮頸癌HeLa細(xì)胞的影響。實(shí)驗(yàn)發(fā)現(xiàn):與單獨(dú)輻照組相比,二硫蘇糖醇聯(lián)合碳離子輻照后細(xì)胞的存活率下降,而凋亡率增加;二硫蘇糖醇聯(lián)合碳離子輻照加重了碳離子輻照引起的細(xì)胞周期阻滯;且聯(lián)合輻照組的自噬被明顯激活。結(jié)果表明,持續(xù)的內(nèi)質(zhì)網(wǎng)應(yīng)激可改變宮頸癌HeLa細(xì)胞對(duì)碳離子輻照反應(yīng),且二硫蘇糖醇可能通過(guò)影響HeLa細(xì)胞的自噬性細(xì)胞死亡通路發(fā)揮作用。
[Abstract]:The effects of endoplasmic reticulum stress on HeLa cells pretreated with different doses of carbon ions were studied. The results showed that the survival rate of cells was decreased and the apoptosis rate was increased after irradiation with dithiothreitol combined with carbon ions, the cell cycle arrest induced by carbon ion irradiation was aggravated by dithiothreitol combined with carbon ion irradiation. Moreover, autophagy in combined irradiation group was significantly activated. The results showed that persistent endoplasmic reticulum stress could change the response of cervical cancer HeLa cells to carbon ion irradiation, and dithiothreitol might play a role in the autophagy cell death pathway of HeLa cells.
