幾丁聚糖抗自然絕經(jīng)大鼠卵巢衰老的作用及機(jī)制

【摘要】:目的探討幾丁聚糖抗自然絕經(jīng)大鼠卵巢衰老的作用及機(jī)制。方法 30只自然絕經(jīng)大鼠模型隨機(jī)分成空白對(duì)照組、實(shí)驗(yàn)組和雌激素組各10只。實(shí)驗(yàn)組給予幾丁聚糖1 500 mg/kg灌胃、雌激素組給予雌二醇(E2)1.2 mg/kg灌胃、空白對(duì)照組給予生理鹽水10 ml/kg灌胃。每天1次,共1個(gè)月。采用酶聯(lián)免疫法測(cè)定大鼠血清中孕激素(P)、E2和白細(xì)胞介素(IL)-2、IL-6、腫瘤壞死因子(TNF)-α水平,黃嘌呤氧化酶法測(cè)定卵巢組織丙二醛(MDA)活力,硫代巴比妥酸法測(cè)定卵巢組織超氧化物歧化酶(SOD)的含量。結(jié)果 1實(shí)驗(yàn)組E2和P高于空白對(duì)照組,實(shí)驗(yàn)組E2低于雌激素組(P均0.05);2實(shí)驗(yàn)組血清IL-2水平明顯高于空白對(duì)照組,而IL-6、TNF-α水平明顯低于空白對(duì)照組(P均0.05)。3實(shí)驗(yàn)組和雌激素組SOD與空白對(duì)照組相比有升高的趨勢(shì),但實(shí)驗(yàn)組低于雌激素組(P0.05);實(shí)驗(yàn)組和雌激素組MDA低于空白對(duì)照組(P0.05)。結(jié)論幾丁聚糖可以通過免疫調(diào)節(jié)作用和抗氧化作用改善卵巢的內(nèi)分泌功能,延緩卵巢衰老。
[Abstract]:Objective to investigate the effect and mechanism of chitosan on ovarian aging in natural menopausal rats. Methods Thirty natural menopausal rats were randomly divided into control group (n = 10) and estrogen group (n = 10). The experimental group was given 1 500 mg/kg of chitosan, the estrogen group 1. 2 mg/kg of estradiol, and the control group 10 ml/kg of normal saline. Once a day for 1 month. The levels of serum progesterone E _ 2 and interleukin (IL) -2 IL-6 and tumor necrosis factor TNF- 偽 were measured by enzyme-linked immunosorbent assay (Elisa), and the activity of malondialdehyde (MDA) in ovarian tissue was determined by xanthine oxidase method. The content of superoxide dismutase (SOD) in ovarian tissue was determined by thiobarbituric acid method. Results 1the levels of serum IL-2 in the experimental group were significantly higher than those in the blank control group, and the levels of serum IL-2 in the experimental group were significantly higher than those in the control group. However, the level of IL-6 TNF- 偽 was significantly lower than that of control group (P < 0.05). The SOD of experimental group and estrogen group was significantly higher than that of control group, but the level of MDA in experimental group and estrogen group was lower than that in control group (P 0.05), but the level of MDA in experimental group and estrogen group was lower than that in control group (P 0.05). Conclusion chitosan can improve the endocrine function of ovary and delay ovarian senescence through immunomodulation and antioxidation.
