【摘要】：為了系統(tǒng)地研究卵巢癌病變過程的信號轉(zhuǎn)導(dǎo)模式,綜合多種信息構(gòu)建了完整的人類信號轉(zhuǎn)導(dǎo)網(wǎng)絡(luò),發(fā)現(xiàn)卵巢癌相關(guān)基因在網(wǎng)絡(luò)中的拓?fù)鋵W(xué)性質(zhì)與其它基因存在顯著差異。將正常、早期卵巢癌和晚期卵巢癌三種狀態(tài)下的基因表達(dá)譜數(shù)據(jù)映射到信號轉(zhuǎn)導(dǎo)網(wǎng)絡(luò)中,利用基于共表達(dá)改變量的方法,篩選出正常與早期卵巢癌、正常與晚期卵巢癌、早期卵巢癌與晚期卵巢癌間的顯著差異模體。通過對顯著差異模體進(jìn)行功能和模式的分析,發(fā)現(xiàn)卵巢癌的發(fā)生發(fā)展主要和信號級聯(lián)模式的異常相關(guān)。另外,還發(fā)現(xiàn)AR、PAK6和KPNB1三個(gè)基因間信號轉(zhuǎn)導(dǎo)模式的異常和卵巢癌的惡化相關(guān)。
[Abstract]:In order to systematically study the signal transduction modes in the process of ovarian cancer, a complete human signal transduction network was constructed by synthesizing many kinds of information. It was found that the topological properties of ovarian cancer related genes in the network were significantly different from those of other genes. The gene expression profiles of normal, early and advanced ovarian carcinomas were mapped to signal transduction networks, and normal and early ovarian carcinomas, normal and advanced ovarian carcinomas were screened by coexpression alteration method. Significant difference motifs between early and advanced ovarian cancer. By analyzing the function and pattern of significantly different motifs, it is found that the occurrence and development of ovarian cancer is mainly related to the abnormal signal cascade pattern. It was also found that the abnormal signal transduction patterns of ARN PAK6 and KPNB1 were associated with the deterioration of ovarian cancer.
