缺氧微環(huán)境下HIF-1α對卵巢癌A2780細(xì)胞侵襲轉(zhuǎn)移的影響及分子機(jī)制

【摘要】:目的:探討缺氧微環(huán)境下HIF-1α在卵巢癌細(xì)胞系A(chǔ)2780侵襲轉(zhuǎn)移中的作用。方法:常氧及缺氧微環(huán)境培養(yǎng)卵巢癌A2780細(xì)胞;RT-PCR及Western blot法檢測缺氧誘導(dǎo)因子-1α(HIF-1α)和基質(zhì)金屬蛋白酶13(MMP13)mRNA及蛋白的表達(dá);siRNA干擾HIF-1α表達(dá),檢測干擾效率及MMP13表達(dá)的變化;Transwell小室侵襲實(shí)驗(yàn)檢測siRNA干擾前后卵巢癌細(xì)胞侵襲轉(zhuǎn)移能力。結(jié)果:缺氧誘導(dǎo)卵巢癌A2780細(xì)胞HIF-1α及MMP13表達(dá)上調(diào),細(xì)胞侵襲數(shù)明顯增多;siRNA能有效抑制HIF-1α表達(dá)及缺氧引起的MMP13增多,降低細(xì)胞侵襲能力。結(jié)論:HIF-1α通過調(diào)控MMP13表達(dá)增強(qiáng)缺氧環(huán)境下卵巢癌細(xì)胞的侵襲能力。
[Abstract]:Objective: to investigate the role of HIF-1 偽 in invasion and metastasis of ovarian cancer cell line A 2780 in hypoxic microenvironment. Methods: RT-PCR and Western blot were used to detect the expression of hypoxia inducible factor-1 偽 (HIF-1 偽) and matrix metalloproteinases (13(MMP13)mRNA) and matrix metalloproteinases (MMP) in A2780 cells cultured in normoxic and hypoxic microenvironment. To detect the interference efficiency and the change of MMP13 expression, the invasive ability of ovarian cancer cells before and after siRNA interference was detected by transwell chamber invasion assay. Results: hypoxia induced upregulation of HIF-1 偽 and MMP13 in A2780 cells, and the increase of cell invasion could effectively inhibit the expression of HIF-1 偽 and the increase of MMP13 induced by hypoxia, and decrease the invasion ability of A2780 cells. Conclusion the expression of MMP13 can enhance the invasion ability of ovarian cancer cells in hypoxic environment by controlling the expression of MMP13 at 1: HIF-1 偽.
