HSP70在不同應(yīng)激程度CUMS昆明小鼠胚胎中的表達(dá)及作用

【摘要】:目的建立雌性昆明小鼠慢性輕度不可預(yù)見性應(yīng)激(CUMS)動(dòng)物模型,探討在心理應(yīng)激狀態(tài)下,熱休克蛋白70(HSP70)是否參與機(jī)體對(duì)胚胎的應(yīng)激保護(hù)作用。方法將300只雌性昆明小鼠以9種應(yīng)激因子在28d內(nèi)隨機(jī)應(yīng)用造模,并分為輕度、中度和重度應(yīng)激3組。對(duì)照組(50只)不造模。通過孕馬血清促性腺激素/人絨毛膜促性腺激素促排卵,觀察卵巢反應(yīng)及胚胎發(fā)育潛能;采用免疫熒光染色及實(shí)時(shí)熒光定量PCR檢測(cè)HSP70在2-細(xì)胞胚胎及孕4d(D4)胚胎中的表達(dá)。結(jié)果 28dCUMS刺激后,實(shí)驗(yàn)組小鼠輕度應(yīng)激占50%(150只),中度應(yīng)激占32%(96只),重度應(yīng)激占18%(54只)。促排卵后,輕度應(yīng)激組小鼠2-細(xì)胞胚胎數(shù)、D4胚胎數(shù)和優(yōu)胚率等與對(duì)照組比較差異均無統(tǒng)計(jì)學(xué)意義,而中、重度應(yīng)激組小鼠較對(duì)照降低(P0.05)。免疫熒光染色和實(shí)時(shí)熒光定量PCR顯示HSP70在輕、中度應(yīng)激組小鼠2-細(xì)胞胚胎和D4胚胎中的表達(dá)高于對(duì)照組(P0.05),而重度應(yīng)激組較對(duì)照組差異無統(tǒng)計(jì)學(xué)意義。結(jié)論 CUMS刺激后HSP70的表達(dá)升高,可能與其應(yīng)激保護(hù)作用有關(guān)。
[Abstract]:Objective to establish the animal model of chronic mild unpredictable stress (CUMS) in female Kunming mice and to explore whether heat shock protein 70 (HSP70) participates in the protective effect of heat shock protein 70 (HSP70) on embryo under psychological stress. Methods 300 female Kunming mice were randomly treated with 9 stress factors within 28 days and divided into three groups: mild moderate and severe stress groups. Control group (n = 50). Ovulation was induced by serum gonadotropin / human chorionic gonadotropin (HCG), ovarian response and embryonic development potential were observed, and the expression of HSP70 in 2-cell embryos and 4-day gestational D4 embryos was detected by immunofluorescence staining and real-time fluorescence quantitative PCR. Results after 28dCUMS stimulation, there were 150 mice in the experimental group under mild stress, 3296 rats in the moderate stress group and 54 rats in the severe stress group. After ovulation induction, there was no significant difference in the number of 2-cell embryos and the rate of superior embryos between the mild stress group and the control group, but in the moderate and severe stress group, the P0.05A was decreased in the severe stress group compared with the control group. Immunofluorescence staining and real-time fluorescence quantitative PCR showed that the expression of HSP70 in 2-cell embryos and D4 embryos in mild and moderate stress group was higher than that in control group (P 0.05), but there was no significant difference between severe stress group and control group. Conclusion the increased expression of HSP70 after CUMS stimulation may be related to its protective effect on stress.
