自發(fā)性早產(chǎn)母體外周血轉(zhuǎn)錄組特征的研究

【摘要】:【目的】 (1)通過(guò)回顧性前瞻研究,分析自發(fā)性早產(chǎn)中母體外周血有核細(xì)胞轉(zhuǎn)錄組的分子特征; (2)通過(guò)對(duì)早產(chǎn)基因mRNA表達(dá)譜分析,篩選出差異表達(dá)基因,為研究早產(chǎn)分子機(jī)制提供線索。 【實(shí)驗(yàn)方法】 (1)基因表達(dá)譜分析:通過(guò)檢測(cè)21例原始樣本(10例自發(fā)性早產(chǎn)和11例足月妊娠)的mRNA表達(dá)譜,分析早產(chǎn)組和足月組間的基因表達(dá)特征,對(duì)其數(shù)據(jù)進(jìn)行整合分析; (2)實(shí)時(shí)定量聚合酶鏈反應(yīng)(RT-PCR):根據(jù)基因表達(dá)譜分析結(jié)果,挑選出13個(gè)上調(diào)差異基因,對(duì)其在另外46例獨(dú)立樣本(21例自發(fā)性早產(chǎn)和25例足月妊娠)中的表達(dá)水平進(jìn)行驗(yàn)證。 【結(jié)果】 (1)基因表達(dá)譜分析結(jié)果:早產(chǎn)組中表達(dá)上調(diào)的基因241個(gè),主要參與炎癥、免疫應(yīng)答、細(xì)胞增殖、DNA復(fù)制、DNA代謝、RNA剪切、RNA代謝、染色質(zhì)組裝等生物學(xué)過(guò)程;表達(dá)下調(diào)的基因690個(gè),主要參與細(xì)胞穩(wěn)態(tài)、信號(hào)轉(zhuǎn)導(dǎo)、離子轉(zhuǎn)運(yùn)、核酸分解代謝等生物學(xué)過(guò)程。 (2)RT-PCR結(jié)果:選取差異最大的13個(gè)上調(diào)基因在獨(dú)立樣本中差異表達(dá)分析,,早產(chǎn)組中NF1表達(dá)水平高于足月組(P<0.05,P=0.008),差異具有統(tǒng)計(jì)學(xué)意義;早產(chǎn)組中CAMK2D表達(dá)水平高于足月組(P<0.05,P=0.028),差異具有統(tǒng)計(jì)學(xué)意義。 【結(jié)論】 (1)自發(fā)性早產(chǎn)的發(fā)生過(guò)程可能與相關(guān)基因激活和抑制有關(guān); (2)NF1和CAMK2D基因表達(dá)異?赡軈⑴c早產(chǎn)的病理生理變化,有望成為預(yù)測(cè)早產(chǎn)的候選分子標(biāo)志物。
[Abstract]:[purpose] 1) the molecular characteristics of nucleated cell transcriptome in maternal peripheral blood during spontaneous preterm delivery were analyzed by retrospective prospective study. 2) differential expression genes were screened by analyzing the mRNA expression profile of preterm birth genes, which provided clues for studying the molecular mechanism of preterm labor. [experimental method] (1) Analysis of gene expression profile: mRNA expression profiles of 21 cases of spontaneous preterm labor and 11 cases of term pregnancy were detected, the characteristics of gene expression between preterm and term groups were analyzed, and the data were integrated and analyzed. (2) Real-time quantitative polymerase chain reaction (RT PCR): according to the results of gene expression profiling, 13 up-regulated differentially expressed genes were selected and their expression levels were verified in another 46 independent samples (21 spontaneous preterm births and 25 term pregnancies). [results] Results of gene expression profile analysis: 241 up-regulated genes in preterm labor group were mainly involved in inflammation, immune response, cell proliferation, DNA replication, DNA metabolism, RNA shearing RNA metabolism, chromatin assembly, and 690 down-regulated genes. Mainly involved in cell homeostasis, signal transduction, ion transport, nucleic acid catabolism and other biological processes. RT-PCR results showed that the expression level of NF1 in preterm labor group was higher than that in term group (P < 0.05), and the difference was statistically significant. The expression of CAMK2D in premature delivery group was significantly higher than that in term group (P < 0.05). [conclusion] 1) the process of spontaneous preterm delivery may be related to the activation and inhibition of related genes; Abnormal expression of NF1 and CAMK2D may be involved in pathophysiological changes of preterm delivery and may be a candidate molecular marker for preterm delivery.
