環(huán)巴胺對子宮內(nèi)膜癌HEC-1A細胞存活與凋亡的影響

【摘要】:目的探討環(huán)巴胺(cyclopamine,CYP)對人子宮內(nèi)膜癌HEC-1A細胞存活及凋亡的影響。方法采用0,5,10,20,40μmol/L CYP處理HEC-1A細胞24 h,倒置顯微鏡下觀察細胞形態(tài)改變,CCK-8法檢測細胞增殖,AO/EB雙染法觀察死亡細胞;流式細胞儀AnnexinⅤ-FITC/PI法檢測細胞發(fā)生的凋亡率,Q-PCR法檢測Bax和Bcl-2基因的表達水平。結(jié)果 CYP處理導致HEC-1A細胞形態(tài)發(fā)生明顯改變,且具有CYP濃度依賴性;CCK-8結(jié)果顯示,CYP可顯著抑制HEC-1A細胞的增殖(P0.05);AO/EB染色結(jié)果表明,CYP可誘發(fā)HEC-1A細胞大量死亡;流式細胞術(shù)檢測發(fā)現(xiàn)CYP可導致HEC-1A細胞凋亡,且隨CYP濃度的升高HEC-1A細胞發(fā)生凋亡的比例升高(P0.05);Q-PCR檢測凋亡相關(guān)基因表達。結(jié)果顯示,CYP處理可誘導HEC-1A細胞上調(diào)表達Bax基因,下調(diào)表達Bcl-2基因。結(jié)論 CYP可抑制人子宮內(nèi)膜癌細胞HEC-1A的存活并誘導其發(fā)生凋亡。
[Abstract]:Objective to investigate the effects of cyclopamine Cyp on the survival and apoptosis of human endometrial carcinoma HEC-1A cells. Methods the cells of HEC-1A were treated with #number0# 渭 mol/L CYP for 24 h. The morphologic changes of HEC-1A cells were observed under inverted microscope.
