ΔNp63α基因過表達(dá)對人宮頸癌細(xì)胞SiHa基因表達(dá)譜的影響

【摘要】:目的分析ΔNp63α基因過表達(dá)時(shí)人宮頸癌細(xì)胞SiHa基因表達(dá)譜的變化,篩選受ΔNp63α調(diào)控的潛在靶基因。方法利用慢病毒轉(zhuǎn)染技術(shù)構(gòu)建穩(wěn)定過表達(dá)ΔNp63α的轉(zhuǎn)染組細(xì)胞株SiHa-ΔNp63α和對照組細(xì)胞株SiHa-NC。應(yīng)用基因芯片技術(shù)研究兩組細(xì)胞株基因表達(dá)譜變化,并進(jìn)行生物學(xué)信息分析和實(shí)時(shí)熒光定量PCR驗(yàn)證。結(jié)果基因芯片數(shù)據(jù)共篩選出差異倍數(shù)1.5倍以上的基因共1 405個(gè),其中843個(gè)基因在SiHa-ΔNp63α細(xì)胞中表達(dá)上調(diào),562個(gè)表達(dá)下調(diào)。通過生物信息學(xué)分析顯示這些基因主要參與細(xì)胞生長與周期、信號轉(zhuǎn)導(dǎo)、細(xì)胞通訊、細(xì)胞黏附、細(xì)胞轉(zhuǎn)移和侵襲等功能,涉及的信號通路包括抗原加工提呈、細(xì)胞因子與受體相互作用、細(xì)胞黏附分子、補(bǔ)體系統(tǒng)等。結(jié)論研究ΔNp63α調(diào)控的潛在靶基因或信號通路,對探索宮頸癌發(fā)生發(fā)展的機(jī)制有重要意義。
[Abstract]:Objective to analyze the changes of SiHa gene expression profile in human cervical cancer cells during overexpression of 螖 Np63 偽 gene, and to screen potential target genes regulated by 螖 Np63 偽.Methods the cell line SiHa- 螖 Np63 偽 of stable overexpression of 螖 Np63 偽 and the control cell line SiHa-NCwere constructed by lentivirus transfection technique.The gene expression profiles of two groups of cell lines were studied by gene chip technique, and the biological information analysis and real-time fluorescence quantitative PCR verification were performed.Results A total of 1 405 genes were screened by microarray data, in which 843 genes were up-regulated and down-regulated in SiHa- 螖 Np63 偽 cells.Bioinformatics analysis showed that these genes were mainly involved in cell growth and cell cycle, signal transduction, cell communication, cell adhesion, cell metastasis and invasion.Cytokines interact with receptors, cell adhesion molecules, complement systems, and so on.Conclusion it is important to study the potential target gene or signal pathway regulated by 螖 Np63 偽 in the pathogenesis of cervical cancer.
