【摘要】：目的探討miR-23和孕激素受體(PR)蛋白在子宮肌瘤組織中的表達(dá)及臨床意義。方法分別用realtimePCR法及western blot法對2013年12月至2014年6月中國醫(yī)科大學(xué)附屬盛京醫(yī)院30例子宮肌瘤組織、30例相應(yīng)肌瘤包膜外子宮肌層組織及10例因子宮脫垂行子宮切除術(shù)的子宮肌層組織中的miR-23和PR蛋白進(jìn)行檢測,并用SPSS17.0統(tǒng)計分析軟件對檢測結(jié)果進(jìn)行統(tǒng)計及相關(guān)性分析。結(jié)果 miR-23在相應(yīng)包膜外子宮肌層組及正常子宮肌層組表達(dá)顯著高于子宮肌瘤組織(P0.001)。孕激素受體A(PRA)、孕激素受體B(PRB)蛋白在相應(yīng)包膜外子宮肌層組及正常子宮肌層組表達(dá)顯著低于子宮肌瘤組織(P0.001)。比較相應(yīng)包膜外子宮肌層組與正常子宮肌層組中PRA、PRB蛋白的相對含量,PRA相對含量差異無統(tǒng)計學(xué)意義(P=0.198),PRB相對含量差異有統(tǒng)計學(xué)意義(P=0.035)。在子宮肌瘤組織中,PRA、PRB蛋白表達(dá)差異無統(tǒng)計學(xué)意義(P=0.389)。miR-23與PRA蛋白表達(dá)之間存在負(fù)相關(guān)關(guān)系(r=-0.754),與PRB蛋白相對含量之間無相關(guān)關(guān)系(r=-0.394)。結(jié)論 miR-23與PR共同促進(jìn)子宮肌瘤的發(fā)生與發(fā)展。PR為miR-23的靶基因之一,miR-23可能通過轉(zhuǎn)錄水平導(dǎo)致PRA蛋白增多。
[Abstract]:Objective to investigate the expression and clinical significance of miR-23 and progesterone receptor protein in uterine leiomyoma.Methods from December 2013 to June 2014, 30 cases of uterine leiomyoma tissues and 10 cases of uterine myometrium tissues of Shengjing Hospital affiliated to China Medical University from December 2013 to June 2014 were treated with realtimePCR method and western blot method respectively, and 10 cases were treated with hysterectomy due to uterine prolapse.The miR-23 and PR proteins in the myometrium of the uterus were detected.SPSS17.0 statistical analysis software was used to analyze the correlation and statistics of the test results.Results the expression of miR-23 in the corresponding extracapsular myometrium group and normal uterine myometrium group was significantly higher than that in uterine myoma tissue (P 0.001).The expression of progesterone receptor Agna and progesterone receptor Bu PRB protein in the corresponding extracapsular myometrium group and normal uterine myometrium group was significantly lower than that in uterine leiomyoma tissue (P 0.001).There was no significant difference in the relative content of PRA between the corresponding extrauterine myometrium group and the normal myometrium group.There was no significant difference in the expression of PRB protein in uterine leiomyoma. There was a negative correlation between the expression of PRA protein and the expression of PRB protein. There was no correlation between PRB protein and the relative content of PRB protein.Conclusion miR-23 and PR can promote the occurrence and development of uterine leiomyoma. PR is one of the target genes of miR-23, which may lead to the increase of PRA protein through transcription level.
