【摘要】：目的通過觀察宮頸上皮內(nèi)瘤變(CIN)和宮頸癌患者外周血、局部組織中芳香烴受體(AhR)的表達,探討AhR與CIN和宮頸癌的關(guān)系,為開拓CIN和宮頸癌的治療新途徑提供理論和實驗依據(jù)。方法選取2010年6月1日-2013年6月1日上海市浦東新區(qū)人民醫(yī)院和上海交通大學(xué)醫(yī)學(xué)院附屬仁濟醫(yī)院的門診和住院患者共140例,因子宮肌瘤、子宮肌腺癥、功能障礙性子宮出血等各種子宮良性病變而行子宮切除,經(jīng)病理學(xué)檢查證實,其中宮頸組織正常者30例(對照組)、CIN患者65例(其中CINⅡ39例和CINⅢ26例,CIN組)、宮頸癌患者45例(宮頸癌組),采用免疫組織化學(xué)和Western印跡法檢測各組患者局部組織中AhR的表達。結(jié)果免疫組織化學(xué)檢測結(jié)果顯示,3組患者局部組織中的AhR陽性信號均位于細(xì)胞質(zhì)和細(xì)胞核。宮頸癌組的AhR指數(shù)為85.56±5.79,顯著高于CIN組的57.25±4.71(t=28.18,P0.05);CIN組的AhR指數(shù)顯著高于對照組的2.29±0.52(t=63.48,P0.05)。Western印跡法檢測結(jié)果顯示,各組局部組織中均有AhR蛋白表達,隨著CIN向?qū)m頸癌的進展,AhR蛋白表達量逐漸增多。宮頸癌組的AhR蛋白相對灰度值為74.96±6.28,顯著高于CIN組的58.33±7.65(t=12.04,P0.05);CIN組的AhR蛋白相對灰度值顯著高于對照組的14.40±6.00(t=27.75,P0.05)。結(jié)論 AhR參與了CIN和宮頸癌的發(fā)生、發(fā)展,AhR可能為CIN和宮頸癌的治療提供新的靶點。
[Abstract]:Objective to investigate the relationship between AhR and CIN and cervical cancer by observing the expression of AhR in peripheral blood and local tissues of cervical cancer patients with cervical intraepithelial neoplasia (cin). To provide theoretical and experimental evidence for the development of new approaches to the treatment of CIN and cervical cancer. Methods from June 1st 2010 to June 1st 2013, Shanghai Pudong New District people's Hospital and Shanghai Jiaotong University School of Medicine were selected.
