老年SD大鼠腦組織間液內(nèi)MR示蹤劑擴散特征的研究

【摘要】:目的隨著腦組織衰老的進展,神經(jīng)細(xì)胞代謝及分泌產(chǎn)物在腦細(xì)胞外間隙(ECS)轉(zhuǎn)運及引流特性發(fā)生不可逆性改變,本研究通過MR示蹤劑在ECS內(nèi)擴散的特征反映老年SD大鼠腦組織間液流動的改變,并通過軟件計算將其量化。材料與方法分別選擇實驗組老年SD大鼠(15~17個月)8只及對照組正常成年SD大鼠(7~10個月)15只,尾狀核注射MR示蹤劑釓噴酸葡胺,分別于0.25 h、0.5 h、1 h、2 h、3 h、4 h進行MRI掃描,觀察造影劑在尾狀核的動態(tài)分布,并測量造影劑的擴散及清除速率。結(jié)果實驗組SD大鼠的示蹤劑在ECS內(nèi)的擴散率D*為(3.25±0.46)×10-4 mm2/s,與對照組的(3.32±0.70)×10-4 mm2/s比較,差異無統(tǒng)計學(xué)意義(t=1.739,P0.05);但實驗組尾狀核區(qū)的示蹤劑清除速率k'為(0.29±0.08)×10-4/s,較對照組的(0.62±0.12)×10-4/s明顯減低,差異有統(tǒng)計學(xué)意義(t=11.602,P0.05)。結(jié)論SD大鼠腦衰老組織的退行性改變引起ECS結(jié)構(gòu)等變化,使腦組織間液清除減慢,這可能與代謝產(chǎn)物蓄積并最終誘發(fā)多種老年性疾病有密切的關(guān)系。
[Abstract]:Objective with the development of brain senescence, the characteristics of (ECS) transport and drainage of neuronal metabolism and secretion products in the extracellular space of brain cells were irreversely changed. In this study, the characteristics of diffusion of MR tracer in ECS were used to reflect the changes of fluid flow in brain tissue of aged SD rats, and the changes were quantified by software calculation. Materials and methods eight aged SD rats (15 ~ 17 months) in the experimental group and 15 normal adult SD rats (7 ~ 10 months) in the control group were selected. The caudate nucleus was injected with MR tracer gadolinium gluconate at 0.25 h, 0.5 h, 1 h, 2 h, respectively, and the control group was treated with MR tracer Gadolinium gluconate at 0.25 h, 0.5 h, 1 h, 2 h, respectively. The dynamic distribution of the contrast agent in the caudate nucleus was observed by MRI scanning at 3 h and 4 h, and the diffusion and clearance rate of the contrast agent were measured. Results the diffusivity of tracer in ECS of SD rats in experimental group was (3.25 鹵0.46) 脳 10 脳 4 mm2/s, and (3.32 鹵0.70) 脳 10 渭 4 mm2/s in control group, there was no significant difference (t = 1.739, P0.05). However, the tracer clearance rate of the caudate nucleus in the experimental group was (0.29 鹵0.08) 脳 10 ~ (4) 渭 s, which was significantly lower than that in the control group (0.62 鹵0.12) 脳 10 ~ 4 / s) (t = 11.602, P0.05). Conclusion the degenerative changes of brain senescence tissue in SD rats cause changes in ECS structure and decrease the clearance of fluid between brain tissues, which may be closely related to the accumulation of metabolite and eventually induce a variety of senile diseases.
