新型載水飛薊賓熱敏脂質(zhì)體-微泡Sono Vue復(fù)合體制備及質(zhì)量分析

【摘要】:目的制備載水飛薊賓熱敏脂質(zhì)體-微泡SonoVue復(fù)合體并進行質(zhì)量分析。方法以薄膜旋蒸法制備載水飛薊賓熱敏脂質(zhì)體,然后利用戊二醛偶聯(lián)法將超聲微泡SonoVue與載藥脂質(zhì)體相連接,通過紫外分光光度法檢測熱敏脂質(zhì)體藥物包封率及不同溫度下釋藥特性。通過激光共聚焦顯微鏡檢測載藥脂質(zhì)體與微泡的偶聯(lián)率。結(jié)果載水飛薊賓熱敏脂質(zhì)體-微泡SonoVue復(fù)合體粒徑(2 724±559.9)nm,藥物包封率(71.0±8.3)%,41℃時藥物平均釋放率明顯升高,達到(55.6±7.6)%。偶聯(lián)后,載藥脂質(zhì)體在微泡周圍聚集,熱敏脂質(zhì)體與微泡偶聯(lián)率為(90.0±4.9)%。結(jié)論載水飛薊素?zé)崦糁|(zhì)體-微泡SonoVue復(fù)合體具有良好的偶聯(lián)率、較高的藥物包封率及控溫釋藥特性,為藥物遞送提供一種新的載體。
