相變型納米粒聯(lián)合低強度聚焦超聲體外顯影及腫瘤治療的實驗研究

【摘要】:第一部分液態(tài)氟碳高分子納米粒體外相變及超聲顯影的實驗研究 目的制備一種新型納米級超聲造影劑—液態(tài)氟碳相變型高分子納米粒,觀察其物理特性、體外熱致相變、聲致相變及超聲顯影效果。 方法采用乳化—蒸發(fā)法(單乳化法)制備包裹液態(tài)氟碳(PFP)的MPEG-PLGA納米粒(P-NP),光鏡觀察納米粒形態(tài)分布;檢測粒徑和電位;在顯微鏡上放置加熱板,觀察P-NP的相變情況;體外行低強度聚焦超聲(low intensity focused ultrasound, LIFU)儀輻照后,觀察其超聲顯影效果。 結果所制備的包裹PFP的高子納米粒外觀為乳白色混懸液,形態(tài)規(guī)則,呈球形;平均粒徑為(393.2±40.7)nm,平均電位為(-5.23±8.69)mV;加熱板顯示在42.6℃左右時,光鏡下顯示納米粒開始轉(zhuǎn)變?yōu)槲馀,且隨著溫度的增高,所產(chǎn)生的氣泡逐漸增多;體外行LIFU儀輻照后,,在超聲基波和諧波模式下可觀察到顯影明顯增強。 結論成功制備的液態(tài)氟碳高分子納米粒粒徑小,穩(wěn)定性好,體外經(jīng)LIFU儀輻照后可增強超聲顯影,其有望成為一種新型的超聲造影劑。 第二部分載藥相變型納米粒聯(lián)合低強度聚焦超聲治療乳腺癌的體外實驗研究 目的研究載紫杉醇相變型納米造影劑聯(lián)合低強度聚焦超聲在輔助化療方面的應用。 方法采用單乳化法制備在紫杉醇相變型納米粒,以掃描及透射電鏡觀察其形態(tài)分布;在顯微鏡下觀察熱相變;于體外應用LIFU輻照后觀察聲相變的超聲顯影;高效液相色譜檢測載藥量、包封率及體外藥物緩釋;臺盼藍檢測LIFU對MCF-7細胞的殺傷作用;CCK-8實驗評估細胞增殖率。 結果掃描電鏡PT-NP呈球形,大小較均勻,表面孔洞樣及毛刷樣結構;透射電鏡PT-NP呈球形,表面毛刷樣結構明顯;體外加熱板43℃時,PT-NP開始相變;聲功率密度0.78w/cm2輻照2min可使PT-NP相變并顯影;PT-NP加LIFU輻照組在48h內(nèi)體外藥物緩釋量最高;LIFU聲功率密度0.78w/cm2輻照2min本身對細胞無明顯毒性作用;體外PT-NP加LIFU輻照組對MCF-7細胞增殖抑制最明顯。 結論成功制備了載藥相變型納米粒,聯(lián)合LIFU輻照,在增強超聲顯影的同時可腫瘤靶向治療。
[Abstract]:Experimental study on phase transition and ultrasonic imaging of liquid fluorocarbon polymer nanoparticles in vitro Aim to prepare liquid fluorocarbon phase change polymer nanoparticles as a new ultrasonic contrast agent, and to observe their physical properties, thermal phase transition in vitro, acoustic phase transition and ultrasonic imaging effect. Methods MPEG-PLGA nanoparticles coated with liquid fluorocarbon (PFP) were prepared by emulsification-evaporation method (single emulsification method). The morphologic distribution of the nanoparticles was observed by light microscope, the particle size and potential were detected, and the phase change of P-NP was observed by placing a heating plate on the microscope. The effect of low intensity focused ultrasound (LIFU) on ultrasound was observed after irradiation with low intensity focused ultrasound (LIFU) in vitro. Results the appearance of the high particle coated with PFP was milky white suspension, regular shape and spherical shape, the average particle size was 393.2 鹵40.7 nm, the average potential was -5.23 鹵8.69 mV.The heating plate showed that at about 42.6 鈩
