靶向超聲造影劑和普通超聲造影劑對(duì)卵巢癌新生血管的評(píng)價(jià)

【摘要】:目的:本研究通過(guò)制備攜血管內(nèi)皮生長(zhǎng)因子受體2單克隆抗體(VEGFR2)的靶向超聲造影劑(MBt),通過(guò)對(duì)比普通超聲造影劑(MBc)與MBt對(duì)裸鼠卵巢癌SKOV3移植瘤新生血管的超聲造影顯像,探討兩種造影劑對(duì)裸鼠卵巢癌移植瘤新生血管的應(yīng)用價(jià)值。方法:裸鼠卵巢癌SKOV3細(xì)胞株皮下移植瘤模型的建立;采用“生物素-親和素橋連接”法制備靶向超聲造影劑;采用免疫熒光法觀察超聲微泡表面VEGFR2單克隆抗體的結(jié)合情況;荷瘤鼠分別注射MBt與MBc行超聲造影檢查并分析靶向組與普通組的時(shí)間強(qiáng)度曲線(TIC)相關(guān)參數(shù);免疫組織化學(xué)檢測(cè)腫瘤組織新生血管的VEGFR2的表達(dá)情況。結(jié)果:成功建立裸鼠卵巢癌SKOV3細(xì)胞株皮下移植瘤模型;免疫熒光法在熒光顯微鏡下觀察到微泡超聲表面發(fā)出的明亮的綠色熒光;靶向組與普通組時(shí)間強(qiáng)度曲線(TIC)分析結(jié)果如下:靶向組的峰值強(qiáng)度73.95(69.50,83.08)dB較普通組57.55(54.30.74.10)dB高(P0.01),靶向組的持續(xù)時(shí)間480.00(442.50,617.50)S較普通組354.00(297.50,412.50)S更長(zhǎng)(P0.01);免疫組織化學(xué)結(jié)果顯示腫瘤組織內(nèi)新生血管的VEGFR2表達(dá)。結(jié)論:攜VEGFR2單克隆抗體的靶向超聲造影劑能夠與裸鼠卵巢癌移植瘤新生血管內(nèi)皮特異性結(jié)合并實(shí)現(xiàn)靶向分子的超聲顯像,故靶向超聲造影技術(shù)對(duì)卵巢癌新生血管形成的評(píng)估提供了新方法。
[Abstract]:Objective: in this study, we prepared the target ultrasound contrast agent of VEGFR2 carrying vascular endothelial growth factor receptor 2 (VEGFR2), and compared the conventional ultrasound contrast agent (MBC) with that of MBt in detecting the neovascularization of SKOV3 transplanted tumor of ovarian cancer in nude mice. To evaluate the value of two contrast agents in the treatment of ovarian cancer xenografts in nude mice. Methods: the model of xenograft tumor in nude mice ovarian cancer SKOV3 cell line was established, and the targeted ultrasound contrast agent was prepared by "biotin-affinity bridge connection" method. Immunofluorescence method was used to observe the binding of monoclonal antibody to VEGFR2 on the surface of ultrasound microbubbles, the tumor bearing mice were injected with MBt and MBc, respectively, and the time intensity curves of target group and normal group were analyzed. The expression of VEGFR2 in tumor tissue was detected by immunohistochemistry. Results: the model of subcutaneous transplantation of ovarian cancer SKOV3 cell line in nude mice was successfully established. The bright green fluorescence from the ultrasonic surface of microbubbles was observed under the fluorescence microscope by immunofluorescence method. The time intensity curves of target group and normal group were as follows: the peak intensity of target group was 73.95 ~ 69.50 ~ 83.08 dB higher than that of normal group (57.55 ~ 54.30.74.10 dB), and the duration of target group was 480.00N 442.50 ~ 617.50S longer than that of normal group (354.00 ~ 297.50 ~ 412.50), and the result of immunohistochemistry showed that the neoplastic tissue was new in tumor tissue. Conclusion: targeting ultrasound contrast agent with VEGFR2 monoclonal antibody can specifically bind to neovascularization endothelium of ovarian cancer xenografts in nude mice and achieve targeted molecular ultrasound imaging. Therefore, targeted contrast-enhanced ultrasonography provides a new method for the evaluation of neoangiogenesis in ovarian cancer.
