【摘要】:目的:探討注射SPIO-shRNA分子探針不同時間點的荷瘤裸鼠腫瘤區(qū)域組織信號強(qiáng)度變化情況,闡明分子探針活體磁共振成像(MRI)機(jī)制和最佳掃描時間。方法:探針注射劑量為18 mg·kg~(-1);將30只BALB/c荷瘤裸鼠隨機(jī)分為6組,每組5只,分別于探針注射前和注射后12、24、27、30和36h等6個時間點行MRI檢查,并測量裸鼠腫瘤組織及對側(cè)肌肉組織在各時間點的信號強(qiáng)度變化;每次檢查完成后立即處死裸鼠,取出腫瘤組織及肌肉組織行HE及普魯士藍(lán)染色,并在光學(xué)顯微鏡下觀察裸鼠腫瘤組織的形態(tài)表現(xiàn)。結(jié)果:與注射前比較,探針注射后12、24和27h裸鼠腫瘤組織T1WI信號強(qiáng)度均升高(P0.05),12、24、27、30和36h腫瘤組織T2WI和T2*GRE信號強(qiáng)度均降低(P0.05或P0.01);探針注射后27h腫瘤組織T1WI信號強(qiáng)度最高;探針注射后27h腫瘤組織T2WI和T2*GRE信號強(qiáng)度低于注射后12、24、30和36h(P0.01)。與注射前比較,注射后24h時裸鼠對側(cè)肌肉組織信號強(qiáng)度降低(P0.05)。HE染色,裸鼠腫瘤組織結(jié)構(gòu)紊亂,細(xì)胞核異型性較多;普魯士藍(lán)染色,在注射后各時間點上均存在藍(lán)染的Fe顆粒,注射后27h時藍(lán)染Fe顆粒最多。結(jié)論:SPIO-shRNA分子探針能成功靶向裸鼠腫瘤組織,且MRI最佳的掃描時間為探針注射后27h。
[Abstract]:Objective: to investigate the changes of signal intensity in tumor region of nude mice injected with SPIO-shRNA molecular probe at different time points. To elucidate the mechanism and optimal scanning time of molecular probe in vivo magnetic resonance imaging (MRI). Methods: the injection dose of the probe was 18 mg 路kg ~ (-1). Thirty BALB/c bearing nude mice were randomly divided into 6 groups, 5 in each group, and MRI was performed at 6 time points before and after injection. The signal intensity of tumor tissue and contralateral muscle tissue in nude mice at different time points were measured. The nude mice were killed immediately after each examination, and the tumor tissues and muscle tissues were taken out for HE and Prussian blue staining, and the morphological features of tumor tissues were observed under optical microscope. Results: compared with before injection. The signal intensities of T1WI in tumor tissues of nude mice were increased at 12h and 27h after the injection of the probe. The signal intensity of T2WI and T2GRE in tumor tissues decreased at 30 and 36 hours (P 0.05 or P 0.01). The signal intensity of T1WI was the highest in the tumor tissue 27 hours after the injection of the probe. The signal intensity of T2WI and T2GRE in tumor tissue at 27 h after injection was lower than that in 1240 h and 36 h after injection, which was compared with that before injection. 24 hours after injection, the signal intensity of contralateral muscle tissue decreased in nude mice. Prussian blue staining showed the presence of blue-stained Fe particles at all time points after injection. Conclusion the MRI molecular probe can successfully target tumor tissue of nude mice, and the best scanning time of MRI is 27h after injection.
