皮質(zhì)酮對(duì)新生大鼠神經(jīng)元遷移蛋白DCX、LIS1表達(dá)的影響

【摘要】:目的研究皮質(zhì)酮(corticosterone)對(duì)新生大鼠大腦神經(jīng)元遷移蛋白DCX(doublecortin、LIS1(microtubule binding lissencephaly1)表達(dá)的影響。 方法將48只生后(postnatal, PN)7d的Sprague-Dawley大鼠隨機(jī)分成對(duì)照組和皮質(zhì)酮干預(yù)組,皮質(zhì)酮干預(yù)組于每日晨起(8時(shí)-9時(shí))腹腔注射皮質(zhì)酮(1.5mg/kg),每日一次,連續(xù)6天;對(duì)照組腹腔注射等量生理鹽水。每組分別于PN14d、17d和21d選取8只斷頭取腦,用Western Blot技術(shù)及免疫組織化學(xué)染色觀察大鼠大腦皮層及海馬DCX、LIS1表達(dá)的變化。 結(jié)果PN14d、17d和21d時(shí),大腦皮層及海馬CA1-2、CA3-4、DG區(qū)皮質(zhì)酮干預(yù)組DCX免疫組化累計(jì)光密度(AOD)均明顯高于正常對(duì)照組(P㩳.05): Western Blot示皮質(zhì)酮干預(yù)組海馬區(qū)DCX蛋白表達(dá)明顯高于正常對(duì)照組(P0.05)。與對(duì)照組比較,日齡14天大腦皮層及海馬CA1-2、CA3-4、DG區(qū)皮質(zhì)酮干預(yù)組LIS1免疫組化AOD均明顯低于正常對(duì)照組(P0.05);日齡14天、21天皮層及日齡21天海馬皮質(zhì)酮干預(yù)組LIS1蛋白表達(dá)明顯低于正常對(duì)照組(P0.05)。 結(jié)論1.LIS1、DCX在正常新生大鼠大腦中表達(dá)隨時(shí)間進(jìn)展表現(xiàn)出一定的規(guī)律性。2.外源性糖皮質(zhì)激素影響神經(jīng)移行相關(guān)蛋白DCX和LIS1表達(dá),可能參與到GC對(duì)腦發(fā)育的影響中。
[Abstract]:Objective to study the effect of corticosterone on the expression of brain neuron migration protein DCX doublecortin LIS1 microtubule binding lissencephaly1 in neonatal rats.Methods 48 postnatal Sprague-Dawley rats of postnatal PN)7d were randomly divided into control group and corticosterone intervention group. In the corticosterone intervention group, 1.5 mg / kg corticosterone was injected intraperitoneally from 8 to 9 am daily for 6 days, while the control group was intraperitoneally injected with the same amount of normal saline.In each group, 8 decapitated rats were selected at 14 d and 21 d, respectively. The changes of the expression of DCXT LIS1 in rat cerebral cortex and hippocampus were observed by Western Blot technique and immunohistochemical staining.Compared with the control group,The immunohistochemical AOD of LIS1 in cortical cortex and hippocampal CA1-2CA3-4DG intervention group was significantly lower than that in normal control group (P 0.05), and the expression of LIS1 protein in cortex and hippocampal corticosterone intervention group was significantly lower than that in normal control group.Conclusion 1. The expression of LIS1 and DCX in the brain of normal neonatal rats showed a certain regularity over time. 2.Exogenous glucocorticoids affect the expression of DCX and LIS1, which may be involved in the effects of GC on brain development.
