【摘要】：研究STAT3突變的常染色體顯性遺傳高lgE綜合征(autosomal dominant hyper-IgE syndrome,AD-HIES)患兒外周循環(huán)B淋巴細胞亞群變化,為研究STAT3基因在B淋巴細胞分化中的作用提供理論基礎。采用多色熒光流式細胞術檢測8例有明確STAT3基因診斷的AD-HIES患兒及10例正常兒童外周循環(huán)B淋巴細胞分化及亞群,并利用統(tǒng)計學方法分析兩組B淋巴細胞亞群的差異。在骨髓及外周B淋巴細胞分化過程中,與同齡對照組相比,AD-HIES患兒外周循環(huán)初始B淋巴細胞顯著增高(P=0.008),而記憶性B淋巴細胞顯著降低(P0.001),其差異均存在統(tǒng)計學意義。在記憶性B淋巴細胞相關亞群中,與同齡正常對照組相比,AD-HIES患兒外周循環(huán)未類別轉化及類別轉化記憶性B淋巴細胞均顯著降低(P=0.002;P=0.001),其差異均存在統(tǒng)計學意義。以上結果說明STAT3基因在外周血B淋巴細胞分化中起著正向調(diào)控作用。
[Abstract]:Study on STAT3 mutation of autosomal dominant lgE syndrome (autosomal dominant hyper-IgE syndrome, AD-HIES) in the peripheral circulation changes of B lymphocyte subsets, provide a theoretical basis for the study of the role of STAT3 gene in B lymphocyte differentiation. By multi-color flow cytometry in 8 AD-HIES patients with clear diagnosis and STAT3 gene 10 cases of normal children's peripheral circulation and the differentiation of B lymphocyte subsets, and the use of statistical methods for analysis of B lymphocyte subsets in two groups. The bone marrow and peripheral B lymphocyte differentiation, compared with age-matched control group, AD-HIES in peripheral B lymphocytes increased significantly in the initial cycle (P=0.008), and memory B cell significantly decreased (P0.001), the differences were statistically significant. Correlation in memory B lymphocyte subsets, compared with normal control group, AD-HIES in peripheral circulation No classification transformation and class transformation memory B lymphocytes were significantly decreased (P=0.002, P=0.001), the difference was statistically significant. The above results showed that STAT3 gene played a positive role in the regulation of B lymphocyte differentiation in peripheral blood.
