【摘要】：目的通過(guò)檢測(cè)DNA甲基轉(zhuǎn)移酶(DNMTs)在兒童免疫性血小板減少癥(ITP)外周血中蛋白水平的表達(dá),探討其與兒童ITP的發(fā)病機(jī)制的關(guān)系。方法收集2015年5月-2016年5月在新鄉(xiāng)醫(yī)學(xué)院第一附屬醫(yī)院門(mén)診、病房就診和治療的60例新診斷ITP患兒(實(shí)驗(yàn)組)和門(mén)診健康體檢的36例兒童(對(duì)照組)為研究對(duì)象。無(wú)菌采集其空腹外周血2 mL,采取酶聯(lián)免疫吸附法檢測(cè)血清中DNMT1、DNMT3a、DNMT3b的蛋白表達(dá)水平。分別分析年齡、性別、血小板計(jì)數(shù)(PLT)對(duì)血清DNMT1、DNMT3a、DNMT3b的影響,及DNMT1、DNMT3a、DNMT3b三者之間的關(guān)系。結(jié)果 (1)新診斷ITP患兒PLT為(21.3±26.1)×10~9/L,明顯低于健康對(duì)照組(261.2±78.4)×10~9/L,差異有顯著性(t=-17.777,P=0.000)。(2)ITP患兒DNMT1、DNMT3a、DNMT3b(ug/L)的蛋白表達(dá)水平分別為(1.43±0.96)、(0.67±0.39)、(0.53±0.39),明顯低于對(duì)照組(2.05±1.24)、(1.04±0.58)、(0.73±0.45),差異有顯著性(t=-2.547,-3.692,-2.341;P=0.013,0.000,0.021)。經(jīng)Person相關(guān)性分析,ITP患兒血小板計(jì)數(shù)與DNMT1、DNMT3a、DNMT3b的蛋白表達(dá)水平無(wú)明顯相關(guān)性(r=-0.062、0.161、0.079,P=0.636、0.220、0.549)。DNMT1與DNMT3a、DNMT3b的蛋白表達(dá)無(wú)相關(guān)性(r=-0.025,-0.057;P=0.851、0.665),DNMT3a、3b的蛋白表達(dá)正相關(guān)性(r=0.259,P=0.046)。結(jié)論新診斷ITP患兒外周血中DNMT1、DNMT3a、DNMT3b的蛋白表達(dá)水平顯著降低,提示DNA甲基轉(zhuǎn)移酶的異常表達(dá)可能與兒童ITP的發(fā)病機(jī)制相關(guān),推測(cè)在兒童ITP患兒DNA甲基化過(guò)程中,DNMT3a、DNMT3b起著相互協(xié)調(diào)的作用。
[Abstract]:Objective to detect the expression of DNA methyltransferase (DNA) in peripheral blood of children with immune thrombocytopenia. Methods from May 2015 to May 2016, we collected the outpatient clinic of the first affiliated Hospital of Xinxiang Medical College. Sixty newly diagnosed children with ITP (experimental group) and 36 healthy children (control group) who were admitted to the ward and treated in the hospital were studied. The fasting peripheral blood samples were collected aseptically and detected by enzyme-linked immunosorbent assay (enzyme-linked immunosorbent assay). The protein expression level of DNMT1, DNMT3a, DNMT3b, DNMT3b, DNMT3b, DNMT3b and DNMT3b. The effect of sex, platelet count and PLT on serum DNMT1, DNMT3aand DNMT3b, Results: the PLT of newly diagnosed children with ITP was 21.3 鹵26.1 脳 10 ~ 9 / L, which was significantly lower than that of the healthy control group (261.2 鹵78.4) 脳 10 ~ (9) / L, and the difference was statistically significant (t -17.777P _ (0.000) P _ (0.000) 路2ITP). Conclusion the expression of DNMT1DNMT3b and DNMT3a DNMT3b in newly diagnosed children with ITP is significantly lower than that of DNMT3a DNMT3b. Conclusion the expression level of DNMT1DNMT3a DNMT3b in newly diagnosed children with ITP is significantly lower than that of DNMT3a DNMT3b, and the positive correlation between DNMT3b and DNMT3a3b protein expression in children with DNMT3aDNMT3b is significantly lower than that of DNMT1DNMT3a DNMT3b protein expression in children with DNMT3aDNMT3b. Conclusion the expression of DNMT1DNMT3a DNMT3b protein in newly diagnosed children with ITP is significantly lower than that of DNMT3a DNMT3b. These results suggest that the abnormal expression of DNA methyltransferase may be related to the pathogenesis of ITP in children, and it is suggested that DNMT3a1 / DNMT3b may play a coordinated role in the process of DNA methylation in children with ITP.
