烏司他丁干預(yù)對膿毒癥幼鼠血清腫瘤壞死因子-α、P-選擇素和凝血酶抗凝血酶復(fù)合物水平的影響

【摘要】:目的探討使用烏司他丁(UTI)作為早期干預(yù)藥物對膿毒癥幼鼠血清腫瘤壞死因子-α(TNF-α)、P-選擇素、凝血酶抗凝血酶復(fù)合物(TAT)水平的影響及意義。方法將120只4周齡雄性大鼠隨機分為正常對照組、假手術(shù)組、膿毒癥組、低劑量UTI干預(yù)組(50 000 U/kg)和高劑量UTI干預(yù)組(200 000 U/kg),每組24只。采用改良的盲腸結(jié)扎穿孔法制作膿毒癥大鼠模型,低劑量和高劑量UTI干預(yù)組在建模后分別尾靜脈注射UTI。ELISA法測定6、12、24 h時各組大鼠血清TNF-α、P-選擇素、TAT水平。結(jié)果膿毒癥組大鼠血清TNF-α、TAT、P-選擇素水平在6 h時即出現(xiàn)顯著升高,血清TNF-α、TAT水平至24 h時仍呈持續(xù)升高狀態(tài)(P0.05);P-選擇素在12 h時達到峰值,24 h時出現(xiàn)回落(P0.05)。UTI干預(yù)組中,P-選擇素和TAT水平變化趨勢和膿毒癥組相同,TNF-α水平在6 h出現(xiàn)明顯的增高,但隨時間延長逐漸降低,且變化幅度均顯著小于膿毒癥組(P0.05);高劑量組變化幅度顯著小于低劑量組(P0.05)。結(jié)論 UTI早期干預(yù)對膿毒癥幼鼠模型的凝血功能有明顯的改善,能抑制TNF-α、P-選擇素、TAT的生成,且高劑量組較低劑量組作用更顯著。
[Abstract]:Objective to explore the effect of ulinastatin (UTI) as an early intervention drug on serum tumor necrosis factor- 偽 (TNF- 偽) and P- selectin in young rats with sepsis. Methods 120 4-week-old male rats were randomly divided into normal control group, sham operation group and sepsis group. Low dose UTI intervention group (50 000 U / kg) and high dose UTI intervention group (200 000 U / kg) were used to make sepsis rat model by modified cecal ligation and perforation method. The serum levels of TNF- 偽 -P- selectin (TNF- 偽 -TNF- 偽 -TATP-selectin) were measured in the low dose and high dose UTI intervention groups at 24 h after modeling, respectively. Results the serum TNF- 偽 -TTATP-selectin level in the septic group increased significantly at 6 h after injection of TNF- 偽 -TATP-selectin. The serum TNF- 偽 tat level continued to increase at 24 h. The level of P0.05 and P- selectin reached its peak at 12 h and reached the peak at 24 h. The change trend of serum TNF- 偽 and TNF- 偽 levels in the intervention group was significantly higher than that in the septic group at 6 h, and the level of TNF- 偽 in the intervention group was significantly higher than that in the septic group at 6 h, and the serum TNF- 偽 level in the intervention group was significantly higher than that in the septic group at 6 h. However, the changes were decreased gradually with the prolongation of time, and were significantly lower than those of sepsis group (P 0.05) and high dose group (P 0.05). Conclusion early intervention of UTI can improve the coagulation function of septic young rats. It could inhibit the production of TNF- 偽 -P- selectin tat, and the effect of high dose group was more obvious than that of low dose group.
