新疆小兒白血病干細(xì)胞免疫表形特點(diǎn)及其相關(guān)研究

【摘要】:目的:測(cè)定維族及漢族患兒白血病干細(xì)胞的表達(dá)水平(Leukemia Stem Cell LSC),檢測(cè)免疫表型、染色體核型,探討其與成人及兩個(gè)民族間是否存在差異,小兒LSC是否存在新的免疫表型,其與染色體核型之間是否存在關(guān)聯(lián)。為白血病個(gè)體化治療提供理論及實(shí)驗(yàn)依據(jù)方法:用流式細(xì)胞儀測(cè)定35例維吾爾族及34例漢族患兒LSC表達(dá)水平,其中選擇10例漢族患兒及所有維族患兒分選其LSC,體外培養(yǎng)觀察其形態(tài)學(xué)特征并再次通過(guò)流式細(xì)胞儀檢測(cè)兩個(gè)民族LSC新的免疫表型,通過(guò)統(tǒng)計(jì)學(xué)方法進(jìn)行兩個(gè)民族CD34+CD38-CD123+細(xì)胞表達(dá)水平之間是否有差異。結(jié)果:兩個(gè)民族AMLLSC的表達(dá)水平在維、漢表達(dá)水平之間存在差異(P=0.04)。培養(yǎng)的LSC呈集落樣懸浮生長(zhǎng),在體外能存活8個(gè)月以上并保持細(xì)胞活性,所檢測(cè)到的表面標(biāo)記有:急性髓系白血病干細(xì)胞(AML LSC)CD34,CD123,CD96,CD44,CD33,CD90.急性淋巴細(xì)胞白血病(ALL-LSC)CD34,CD19,CD9,CD90。CD123+,,CD90+在急淋及急非淋LSC中均有不同程度的表達(dá)。在維吾爾族染色體核型有:ALL t(12;21),現(xiàn)t(9;22),超二倍體等,AML中t(8;21), t(15:17).資料顯示5例M3中4例表達(dá)CD96的LSC都出現(xiàn)t(15:17)核型異常結(jié)論:AMLCD34+CD38-CD123+細(xì)胞表達(dá)水平明顯低于成人,在維吾爾族及漢族白血病患兒存在差異,ALL LSC除了表達(dá)文獻(xiàn)相似的免疫表型外,還表達(dá)了CD34+CD9+,CD34+CD19+等,這些可能小兒LSC新的免疫表型; CD123+,CD90+可能在急淋及急非淋LSC中均存在;CD96表達(dá)可能與t(15:17)異常有關(guān)。
[Abstract]:Objective: to determine the expression level of leukemia stem cells in children of Uygur nationality and Han nationality and to detect the immunophenotype and karyotype of leukemia Cell, and to explore whether there are differences between the leukemia stem cells of Uygur nationality and Han nationality, and whether there is a new immunophenotype of LSC in children. To provide theoretical and experimental basis for individualized treatment of leukemia, flow cytometry was used to determine the expression of LSC in 35 children with Uygur nationality and 34 children with Han nationality. Among them, 10 children of Han nationality and all children of Uygur nationality were selected, their morphological characteristics were observed in vitro and the new immunophenotypes of LSC of two nationalities were detected by flow cytometry. Results: the expression level of AMLLSC in the two nationalities was different in Uygur and Han nationality. The cultured LSC was colony like suspension growth. Can survive for more than 8 months in vitro and maintain cell activity. The surface markers detected were as follows: acute myeloid leukemia stem cell (AMLL) LSC-CD34-CD123ONCD96-CD4C4+ CD33-CD90.Acute lymphoblastic leukemia (ALL-LSCT) CD34T CD34T CD19 CD90.CD90.CD90.CD12123 CD90 was expressed in acute lymphoblastic leukemia (AMLL) and acute non-lymphoid leukemia (LSC) to varying degrees. The chromosome karyotype of Uygur nationality has a chromosome karyotype of: all all 1221. The data showed that 4 cases of LSC expressing CD96 in 5 cases of M3 had abnormal karyotype (t 15: 17) conclusion the expression level of CD38-CD123 cell in the CD38-CD123 cell of CD96 was significantly lower than that in the adult. In Uygur and Han nationality children with leukemia, all LSC expressed CD34 CD9 CD34 CD19, in addition to expressing similar immunophenotypes in literature. These new immunophenotypes of LSC may be related to the abnormal expression of CD123 / CD90 in both acute and non-lymphoid LSC.
