【摘要】：目的了解呼吸道合胞病毒(RSV)感染患兒呼吸道上皮細(xì)胞中干擾素(IFN)-λ1的表達(dá)水平及其與RSV載量的關(guān)系。方法收集2015年6月至2016年6月期間因呼吸道感染住院患兒的鼻咽拭子標(biāo)本,采用直接免疫熒光法行7種常見(jiàn)呼吸道病毒(含RSV)抗原檢測(cè),篩選出單純RSV陽(yáng)性患兒作為RSV感染組(n=120);另收集同期門診體檢健康兒童鼻咽拭子標(biāo)本,行7種常見(jiàn)呼吸道病毒檢查均為陰性的兒童作為健康對(duì)照組(n=50)。對(duì)兩組兒童鼻咽拭子標(biāo)本行熒光定量PCR檢測(cè)RSV載量和IFN-λ1 m RNA水平。結(jié)果 RSV感染組鼻咽拭子中IFN-λ1 m RNA水平顯著高于健康對(duì)照組(P0.05),且IFN-λ1 m RNA水平與RSV載量呈正相關(guān)(r=0.56,P0.05)。結(jié)論 RSV可誘導(dǎo)呼吸道上皮細(xì)胞中IFN-λ1的表達(dá),推測(cè)IFN-λ1在機(jī)體抗RSV感染中可能起重要作用。
[Abstract]:Objective to investigate the expression of IFN- 位 1 in respiratory epithelial cells of children with respiratory syncytial virus (RSV) infection and its relationship with RSV load. Methods the nasopharyngeal swabs of children hospitalized with respiratory tract infection from June 2015 to June 2016 were collected. Seven common respiratory virus (RSVV) antigens were detected by direct immunofluorescence assay, and only RSV positive children were selected as RSV infected group, and nasopharyngeal swabs were collected from healthy children in the same period of outpatient examination. Seven common respiratory virus negative children were used as healthy control group. The RSV load and IFN- 位 1 m RNA level in nasopharynx swab of two groups of children were detected by fluorescence quantitative PCR. Results IFN- 位 1 m in nasopharynx swab of RSV infected group was determined by fluorescence quantitative PCR. The level of RNA was significantly higher than that of control group (P 0.05), and the level of IFN- 位 1 m RNA was positively correlated with the RSV load. Conclusion RSV can induce the expression of IFN- 位 1 in respiratory epithelial cells. It is speculated that IFN- 位 1 may play an important role in RSV infection.
