【摘要】：目的檢測(cè)趨化因子CXCL10及其受體CXCR3在幼年特發(fā)性關(guān)節(jié)炎(JIA)患兒關(guān)節(jié)滑膜組織中的表達(dá),探討其在JIA發(fā)病機(jī)制中的作用。方法通過免疫組織化學(xué)方法,檢測(cè)12例JIA患兒和4例非JIA患兒關(guān)節(jié)滑膜組織中CXCL10和CXCR3的表達(dá);通過半定量RT-PCR方法,檢測(cè)CXCR3在JIA患兒和對(duì)照幼兒關(guān)節(jié)滑膜組織中mRNA水平的表達(dá)。結(jié)果免疫組織化學(xué)結(jié)果表明,CXCL10/CXCR3在JIA患兒及對(duì)照組陽(yáng)性表達(dá)差異有顯著性(P0.05);CXCR3在JIA患兒關(guān)節(jié)滑膜組織中mRNA表達(dá)水平(CXCR3∶GAPDH為2.26±1.55)顯著高于對(duì)照組(0.66±0.44),兩者差異有顯著性(P0.05)。結(jié)論趨化因子CXCL10及其受體CXCR3在幼年特發(fā)性關(guān)節(jié)炎(JIA)的發(fā)病中可能起到了重要的作用。
[Abstract]:Objective to detect the expression of chemokine CXCL10 and its receptor CXCR3 in synovium of children with juvenile idiopathic arthritis (JIA) and to explore the role of them in the pathogenesis of JIA. The expression of CXCL10 and CXCR3 in synovial tissues of 12 children with JIA and 4 children with non JIA were detected, and the expression of CXCL10 and CXCR3 in synovial tissues of 12 children with JIA and 4 children with non JIA were detected. The expression of CXCR3 in synovial tissue of children with JIA and control was detected. Results the positive expression of CXCL10 / CXCR3 in children with JIA and control group was significantly higher than that in control group. There was a significant difference in mRNA expression in synovial tissue of children with JIA. The expression level of CXCR3: GAPDH was 2.26 鹵1.55, which was significantly higher than that of control group (0.66 鹵0.44). Conclusion the chemokine CXCL10 and its receptor CXCR3 may play an important role in the pathogenesis of juvenile idiopathic arthritis.
